摘 要:目的:探讨川芎提取物对脑瘫模型小鼠的干预效果及机制。方法:50只健康昆明种雄性小鼠随机分为正常组、模型组、川芎提取物低、中、高剂量组,每组10只。采用握持牵引试验和足错误试验进行小鼠行为学评价,2,3,5-三苯基氯化四氮唑(TTC)法观察小鼠病理组织,ELISA检测神经生长因子(NGF)水平,RT-PCR检测咬合蛋白mRNA表达。Western blot检测p-环磷腺苷效应元件结合蛋白(CREB)、基质金属蛋白酶-9(MMP-9)、脑源性神经营养因子(BDNF)、p-Akt表达。结果:与模型组相比,川芎提取物低、中、高剂量组小鼠牵引握持时间延长,足错误次数减少(P<0.05),MMP-9、NGF、p-CREB表达降低,p-Akt、BDNF、咬合蛋白表达升高(P<0.05),且呈剂量依赖性。结论:川芎提取物可抑制脑瘫模型小鼠脑损伤,其作用机制可能与CREB信号通路有关,高剂量川芎提取物效果更为显著。Objective:To study intervention effect and mechanisms of Chuanxiong extract on cerebral palsy model mice.Methods:A total of 50 healthy Kunming species male mice were randomly divided into normal group,model group,Chuanxiong extract low,medium and high doses groups,with 10 mice in each group.Behavioral of mice was evaluated by grip traction test and foot error test,and pathological tissues were observed by 2,3,5-triphenyltetrazole chloride(TTC).Nerve growth factor(NGF)level was detected by ELISA,and Occlusal protein mRNA expression was detected by RT-PCR.p-cAMP-response element binding protein(CREB),immunoimprinting matrix metalloproteinase-9(MMP-9),brain-derived neurotrophic factor(BDNF)and p-Akt expressions were detected by Western blot.Results:Compared with model group,holding time of mice was extended and foot errors were reduced in Chuanxiong extract low,medium and high doses groups(P<0.05),MMP-9,NGF and p-CREB expressions were decreased,p-Akt,BDNF and Occludin expressions were increased(P<0.05),which was in a dose-dependent manner.Conclusion:Chuanxiong extract can inhibit brain damage in mice with cerebral palsy,whose mechanism may be related to CREB signaling pathway,and high-dose Chuanxiong extract has more obvious effect.
