摘 要:目的探讨微小RNA(miRNA)-27与宫颈癌细胞线粒体凋亡途径的相关性。方法通过miRNA数据库对miRNA-27进行生物信息学分析,并在体外构建宫颈癌细胞SiHa凋亡模型,采用实时荧光定量聚合酶链反应检测相关分子表达改变。结果miRNA-27存在4种成熟体hsa-miRNA-27a-3p、hsa-miRNA-27a-5p、hsa-miRNA-27b-3p和hsa-miRNA-27b-5p,在宫颈癌组织中表达不一,其靶基因参与多种生物学功能和信号通路,并与线粒体凋亡途径密切相关,其中靶基因LETM1结构域蛋白1(LETMD1)与宫颈癌密切相关并与miRNA-27共表达存在相关性,进一步在宫颈癌细胞SiHa凋亡模型中验证发现,hsa-miRNA-27a-3p、hsa-miRNA-27a-5p表达下调而LETMD1表达升高。结论miRNA-27与宫颈癌细胞线粒体凋亡途径密切相关,可能与LETMD1相关,有望成为潜在的治疗靶点。Objective To investigate the correlation between microRNA(miRNA)-27 and mitochondrial apoptosis pathway in cervical cancer cells.Method The bioinformatics analysis of miRNA-27 was carried out through the miRNA database,the SiHa apoptosis model of cervical cancer cells were constructed in vitro,and the expression changes of related molecules were detected by real-time fluorescent quantitative polymerase chain reaction.Result There were four mature forms of miRNA-27,hsa-miRNA-27a-3p,hsa-miRNA-27a-5p,hsa-miRNA-27b-3p,and hsa-miRNA-27b-5p.They were expressed differently in cervical cancer tissues,and their target genes were involved in various biological functions and signaling pathways,and closely related to the mitochondrial apoptosis pathway.Among them,the target gene LETM1 domain containing 1(LETMD1)was closely related to cervical cancer and correlated with the co-expression of miRNA-27.Further verification in the cervical cancer cell SiHa apoptosis model found that the expression of hsa-miRNA-27a-3p and hsa-miRNA-27a-5p were down-regulated while the expression of LETMD1 was up-regulated.Conclusion miRNA-27 is closely related to the mitochondrial apoptosis pathway of cervical cancer cells and may be related to LETMD1,which is expected to become a promising therapeutic target.
