摘　　要：目的研究肌凝蛋白1E(myosin 1E,MYO1E)在胰腺癌中的表达差异、临床意义及其对生存预后的影响。探究MYO1E过表达对胰腺癌细胞株SW1990细胞生物学功能的影响。方法从癌症基因组图谱(the cancer genome atlas,TCGA)数据库中下载胰腺癌单基因表达信息及患者临床病理特征。用R软件分析MYO1E mRNA在胰腺癌组织及正常胰腺组织中的表达差异。分析MYO1E表达差异与患者临床病理特征及预后的关系。构建MYO1E过表达质粒,将其转染至胰腺癌细胞株SW1990细胞中,并观测MYO1E过表达对胰腺癌细胞株SW1990细胞生物学行为的影响。结果MYO1E mRNA在胰腺癌组织中的表达量高于正常胰腺组织(3.825±0.689 vs.2.788±0.489,t=16.033,P<0.05)。MYO1E表达量与胰腺癌患者的年龄及分化程度密切相关(P均<0.05),并且MYO1E mRNA高表达组患者的生存时间低于MYO1E mRNA低表达组(P<0.05)。单因素Cox回归分析显示,年龄、淋巴结转移和MYO1E表达水平与胰腺癌患者预后相关(P均<0.05)。多因素Cox回归分析显示,年龄、淋巴结转移和MYO1E表达水平是胰腺癌患者预后的独立危险因素(P均<0.05)。细胞计数试剂盒(cell counting kit 8,CCK-8)实验结果显示,MYO1E组24、48 h吸光度值均高于对照组(P均<0.05)。Transwell实验和细胞划痕实验结果显示,MYO1E组侵袭细胞计数均高于对照组(P均<0.05)。流式细胞仪检测结果显示,MYO1E组细胞凋亡率低于对照组(P<0.05)。结论MYO1E在胰腺癌组织中高表达并且其表达水平与患者的预后紧密联系。MYO1E过表达能够促进胰腺癌细胞的增殖,增强其侵袭及转移的能力,并抑制胰腺癌细胞的凋亡。Objective To investigate the expression and clinical significance of myosin 1E(MYO1E)in pancreatic cancer and its effects on survival and prognosis.To explore the effects of MYO1E overexpression on the biological function of pancreatic cancer cell strain SW1990.Methods The single gene expression information and clinicopathological characteristics of pancreatic cancer patients were downloaded from the cancer genome atlas(TCGA)database.The difference of MYO1E mRNA expression between pancreatic cancer tissues and normal pancreatic tissues were analyzed by R software.The relationship between the MYO1E expression difference and the clinicopathological characteristics as well as prognosis of patients was analyzed.The MYO1E overexpression plasmid was constructed and transfected into pancreatic cancer cell strain SW1990,and the effects of MYO1E overexpression on the biological behavior of pancreatic cancer cell strain SW1990 was observed.Results The expression of MYO1E mRNA in pancreatic cancer tissue was higher than that in normal pancreatic tissue(3.825±0.689 vs.2.788±0.489,t=16.033,P<0.05).The expression of MYO1E was closely related to the age and differentiation of pancreatic cancer patients(all P<0.05),and the survival time of patients with high MYO1E mRNA expression was lower than that of patients with low MYO1E mRNA expression(P<0.05).The univariate Cox regression analysis showed that age,lymph node metastasis and MYO1E expression were related to the prognosis of pancreatic cancer patients(all P<0.05).The multivariate Cox regression analysis showed that age,lymph node metastasis and MYO1E expression were independent risk factors for the prognosis of pancreatic cancer patients(all P<0.05).The results of cell counting kit 8(CCK-8)showed that the absorbance values of MYO1E group at 24 and 48 hours were higher than those of the control group(all P<0.05).Transwell test and cell scratch test showed that the number of invasive cells in MYO1E group was higher than that in control group(all P<0.05).The results of flow cy
