摘 要:目的研究1株浙江舟山东海沉积物来源真菌Talaromyces sp.ZHS32次级代谢产物及其抗菌活性。方法利用硅胶柱层析、Sephadex LH-20凝胶柱层析和半制备高效液相色谱等分离方法对该菌株发酵粗提物进行分离纯化;通过核磁共振波谱(NMR)、质谱(MS)等方法并结合相关文献数据比对鉴定化合物结构;采用肉汤微量稀释法对化合物进行抗菌活性评价。结果从海洋真菌Talaromyces sp.ZHS32中分离鉴定了7个已知化合物,分别为dankasterone A(1)、penifupyrone(2)、3-O-methylfunicone(3)、alternariol(4)、rubralactone(5)、orsellinic acid(6)和2-formyl-3,5-dihydroxy-6-methylbenzoic acid(7);体外抗菌活性测试结果表明,化合物1对幽门螺杆菌有较好的抑制作用。结论从海洋真菌Talaromyces sp.ZHS32中分离得到7个化合物,化合物1具有抑制幽门螺杆菌生长的活性。Objective To study the secondary metabolites and their biological activities of the marine-derived fungus Talaromyces sp.ZHS32 isolated from a sediment sample that was collected from the East China Sea,Zhoushan,Zhejiang,P.R.of China.Methods The silica gel column chromatography,Sephadex LH-20 gel column chromatography and semi-preparative high-performance liquid chromatography(HPLC)were used to isolate and purify the secondary metabolites from the fermented extracts of the fungus Talaromyces sp.ZHS32.Their structures were elucidated by modern spectroscopy methods such as nuclear magnetic resonance(NMR)and mass spectrometry(MS)combined with relevant literature data.Their antibacterial activities were evaluated by microdilution method.Results Seven known compounds were obtained from Talaromyces sp.ZHS32,which were identified as dankasterone A(1),penifupyrone(2),3-O-methylfunicone(3),alternariol(4),rubralactone(5),orsellinic acid(6)and 2-formyl-3,5-dihydroxy-6-methylbenzoic acid(7).Compound 1 showed moderate antibacterial activities to Helicobacter pylori.Conclusion Seven compounds(1-7)were isolated from one marine-derived fungus Talaromyces sp.Compound 1 showed antibacterial activities against H.pylori in vitro.
