摘  要:本文研究毛水苏多糖(polysaccharides from Stachys baicalensis,SBP)对糖尿病小鼠肝脏的保护作用。采用高糖饲料喂养结合腹腔注射链脲佐菌素(STZ)诱导建立Ⅱ型糖尿病模型,连续灌胃28d后,考察其对糖尿病小鼠体重、空腹血糖水平、肝脏指数、肝功生化指标的影响,并进行病理学检测,采用免疫组化和qRT-PCR法测定肝脏组织NF-κB、ICAM-1、MCP-1蛋白和mRNA水平。毛水苏多糖可明显减轻糖尿病症状,体重异常情况明显改善,空腹血糖值与肝脏指数明显降低。给药组小鼠肝功生化指标TG、TC、LDL-C、ALT和AST含量均极显著升高,HDL-C含量显著降低,对抑制肝脏代谢紊乱和修复组织损伤肿胀具有良好的效果,有效降低肝脏组织中NF-κB、ICAM-1、MCP-1蛋白和mRNA水平。毛水苏多糖对于STZ诱导的糖尿病小鼠的肝脏保护作用可能与NF-κB介导的炎症通路有关。To explore the protective effect of polysaccharides from Stachys baicalensis(SBP)on liver of diabetic mice,Streptozocin(STZ)induction combined with high-sugar feed was used to establish a diabetic mice model.After continuous intragastric administration of mice for 28 d,the body weight and blood sugar of mice were measured,liver function biochemical indexes,and pathological section of liver were observed.The protein and mRNA level of NF-κB,ICAM-1 and MCP-1 of in liver of diabetic mice was measured by immunohistochemistry and qRT-PCR method.The results showed that SBP could significantly reduce symptoms of diabetes,significantly improve abnormal body weight,and significantly reduce fasting blood glucose level and liver indexes.The contents of liver function biochemical indexes TG,TC,LDL-C,ALT and AST in the administration group were significantly increased,and HDL-C was significantly reduced.SBP could inhibit liver metabolic disorder and had a good effect on repairing tissue damage.It could effectively reduce the protein and mRNA level of NF-κB,ICAM-1 and MCP-1 of in liver of diabetic mice.The conclusion was that the protective effect of SBP on the liver of STZ-induced diabetic disease mice may be related to NF-κB-mediated inflammatory pathways.
