摘  要:目的:探究诺丽果多糖通过对小鼠大脑神经递质和热应激蛋白调节改善机体疲劳机制。方法:将配置的不同浓度诺丽果多糖口服喂养雄性小鼠8周后,采用开敞箱试验(OFT)、强迫游泳试验(FST)和悬尾试验(TST)评价诺丽果多糖的抗疲劳作用,采用增强化学发光法分析热应激蛋白的表达量。结果:OFT试验中,诺丽果多糖10和100 mg/kg组显著增加了小鼠总运动距离(P<0.05),FST和TST试验中,诺丽果多糖10和100 mg/kg组显著减少了小鼠静止时间(P<0.05)。FST试验后小鼠脑干和下丘脑中去甲肾上腺素(norepinephrine,NE)、5-羟色胺(5-hydroxytryptamine,5-HT)、5-羟基吲哚乙酸(5-hydroxyindoleacetic acid,5-HIAA)和多巴胺(dopamine,DA)水平均呈现降低的趋势,其中NE、5-HT和DA降低显著(P<0.05)。诺丽果多糖喂食小鼠心肌HSP70表达量显著升高(P<0.05)。结论:诺丽果多糖通过调节小鼠大脑神经递质和热应激蛋白起到改善疲劳增强运动耐受性的作用,可用于开发新型抗疲劳产品。To investigate Morinda citrifolia L.fruit polysaccharide mechanism of reducing body exercise fatigue by regulating brain neurotransmitters and heat stress proteins in mice.Male mice were fed oral Morinda citrifolia L.fruit polysaccharide of different concentrations for 8 weeks,open field tes(t OFT),forced swimming tes(t FST),and tail suspension test(TST)were used to evaluate the anti-fatigue effects;the expression of heat stress protein was analyzed by enhanced chemiluminescence method.Results In OFT test,10 and 100 mg/kg Morinda citrifolia L.fruit polysaccharide groups,the total exercise distance of mice was significantly increased(P<0.05),in FST and TST tests,10 and 100 mg/kg Morinda citrifolia L.fruit polysaccharide groups,the resting time of mice was significantly reduced(P<0.05).After FST test,levels of norepinephrine(NE),5-hydroxytryptamine(5-HT),5-hydroxyindoleacetic acid(5-HIAA)and dopamine(DA)in mice brain stem and hypothalamus were decreased,and levels of NE,5-HT and DA were significantly decreased(P<0.05).The expression of HSP70 in myocardium of mice fed with Morinda citrifolia L.fruit polysaccharide was significantly increased(P<0.05).Conclusion Morinda citrifolia L.fruit polysaccharide allevated fatigue and enhanced exercise tolerance by regulating brain neurotransmitters and heat stress proteins in mice.It can be used in developing new anti-fatigue products.
