摘  要:以不同时期的枣果实、枣花及枣叶片为材料,采用高效液相色谱法对枣中苯甲酸含量的时空分布进行测定。将枣果果肉烘干、磨粉,按料液比1∶5(g∶mL)的比例加入甲醇提取液,15℃超声提取60 min,过0.22 mm滤膜,在色谱柱为依利特Hypersil BDS C_(18)(4.6 mm×250 mm,5 mm)不锈钢柱,流动相为乙腈:水(pH3.0)体积比15∶85(v/v),柱温35℃,检测波长230 nm,流速1.0 mL/min,进样量10 mL条件下进样,外标峰面积法定量。以期为红枣中苯甲酸含量的理论研究提供参考依据。结果表明:枣叶中不含有苯甲酸或其含量水平极低,枣果中一般只有在果实发育中后期(白熟期、半红期、全红期)才能检测到苯甲酸。所有样品中检测到的苯甲酸含量水平较低,为痕量~7.75 mg/100g DW。在不同时间和地区采集的所有样品中,‘骏枣’的苯甲酸含量最高达8.61 mg/100g DW,其次为‘金丝小枣’,含量最高为4.14 mg/100g,‘灰枣’是苯甲酸含量较少的品种,其苯甲酸含量最高只有3.03 mg/100g DW。The benzoic acid content in jujube was determined by HPLC.The jujube pulp was dried and ground,and then extracted by methanol at ratio of material to liquid 1∶5(g∶mL),and assisted by ultrasonic at 15℃for 60 min.The extraction liquid was then filtered through 0.22 mm filter membrane,and elite hypersil BDS C_(18)(4.6 mm)was used on chromatographic column(4.6 mm×250 mm,5 m)stainless steel column.Mobile phase is acetonitrile:water(pH 3.0),the volume ratio was 15∶85(v/v),the column temperature was 35℃,the detection wavelength was 230 nm,the flow rate was 1.0 mL/min,and injection volume was 10 mL.Sample was quantified by external standard peak area to provide reference for the theoretical study of benzoic acid content in jujube.The results showed that there was no benzoic acid in jujube leaves or its content level was not detectable.Generally,benzoic acid could be detected only in the middle and late stage of fruit development(white ripening stage,half red stage and full red stage).The benzoic acid content detected in all samples was low at trace~7.75 mg/100 g DW.Among all samples collected at different times and regions,‘Jun jujube’has the highest benzoic acid content of 8.61 mg/100 g DW,followed by‘Jinsi jujube’,with the highest content of 4.14 mg/100 g,and‘grey jujube’with the highest benzoic acid content of 3.03 mg/100 g DW.
