摘 要:为了明确玉米须多糖对高尿酸血症小鼠的影响,本试验采用生物信息学和网络药理学的方法推测玉米须对基因干预造模的高尿酸血症模型小鼠的作用机制;使用药物干预造模的高尿酸血症模型小鼠进行体内试验,通过检测小鼠体重、血液生化指标、肾脏指数和肾脏组织病理学检查,验证玉米须多糖对高尿酸血症小鼠肾脏的保护作用。预测结果显示,玉米须主要通过小分子分解代谢、氨基酸代谢、氧化还原酶活动等多种生物学过程以及氨基酸合成、胰岛素抵抗等信号通路对小鼠高尿酸血症产生影响。体内验证试验结果显示,与模型组相比,低、高浓度玉米须多糖组小鼠血清尿素、尿酸和肌酐水平均显著降低(P<0.05或P<0.01),肾脏指数有所降低,但差异不显著(P>0.05),小鼠肾小管的扩张得到不同程度的缓解。结果表明,玉米须多糖可降低高尿酸血症小鼠血清中尿素、尿酸和肌酐水平,并对肾脏组织起到一定的保护作用。本试验结果可为临床高尿酸血症的预防和治疗提供参考。In order to clarify the effect of corn silk polysaccharide on hyperuricemia mice,this study utilized bioinformatics and network pharmacological methods to uncover the mechanisms of corn silk on hyperuricemia mice induced by gene intervention.The protective effect of corn silk polysaccharide on the kidney of hyperuricemia mice was verified by body weight,blood biochemical index,kidney index and kidney histopathology.The predicted results showed that the effects of corn silk on hyperuricemia mice were related mainly to a variety of biological processes,such as small-molecule catabolism,amino acid metabolism,oxidoreductase activity,as well as amino acid synthesis,insulin resistance and other signaling pathways.Furthermore,compared with the model group,the levels of urea,uric acid and creatinine in serum of the mice treated with low and high concentrations of corn silk polysaccharide were significantly decreased(P<0.05 or P<0.01),the kidney index was decreased,but the difference was not significant(P>0.05),and the dilatation of the renal tubules was relieved in different degrees.Thus,in hyperuricemia mice,corn silk polysaccharide significantly reduced serum urea,uric acid and creatinine levels,and had a certain protective effect on kidney tissue.The results of this study can provide reference for the prevention and treatment of clinical hyperuricemia.
