摘 要:基于超高效液相色谱-四极杆-飞行时间质谱(UPLC-Q-TOF/MS)技术研究了绿原酸、莨菪亭和芸香苷3种烟草多酚在正/负离子电喷雾电离模式下形成的加合离子,探讨了不同流动相组成对多酚加合离子形式、丰度和强度的影响。结果表明:(1)在负离子模式下,3种多酚的主要离子形式均为[M-H]-,所占比例超过96%,其他形式加合离子对定量的影响有限,但不同流动相组成对绿原酸和芸香苷加合离子形式以及3种多酚加合离子强度均有一定程度的影响。(2)在正离子模式下,不同流动相组成对多酚加合离子的形式无明显影响,但对多酚加合离子的丰度和强度影响较大,且该影响也与多酚的结构相关。(3)不同加合离子碎裂规律的差异有助于化合物结构的鉴定,能够提高定性分析的准确性。Based on ultra-high performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry(UPLC-Q-TOF/MS),the adduct ions formed by three tobacco polyphenols,including chlorogenic acid,scopoletin and rutoside,were studied in either positive/negative electrospray ionization mode.The effects of mobile phase composition on the form,abundance and intensity of polyphenol adduct ions were investigated.The results showed that:1)In negative ion mode,the major ion form of all the three polyphenols was[M-H]-with an abundance of more than 96%,and thus the formation of the other adduct ions had a limited effect on quantification.However,the mobile phase composition had certain effects on the form of the chlorogenic acid and rutoside adduct ions and the intensities of the adduct ions of the three polyphenols tested.2)In the positive ion mode,the mobile phase composition had no significant effect on the forms of the polyphenol adduct ions.However,it had significant effects on the abundances and intensities of the polyphenol adduct ions,which were also related to the structures of the polyphenols.3)Differences in the fragmentation patterns of different adduct ions were generated and used in identifying the compound structure and could improve the accuracy of qualitative analysis.
