摘  要:目的了解江西省2005-2020年猪链球菌菌株病原学和基因组特征,分析15年间分离株之间的关联,为人感染猪链球菌病防控提供参考依据。方法通过聚合酶链反应法扩增猪链球菌基因组上的16S rRNA、cps2j、mr、sly、ef、gapdh毒力基因,通过测定最小抑菌浓度确定菌株的耐药性,通过全基因组测序和序列比对分析菌株之间的遗传多态性。结果10株人源和8株猪源猪链球菌均为血清2型菌株,毒力基因谱不同,至少携带3种毒力基因。对阿奇霉素和四环素耐药率高达89.89%和77.78%,携带氨基糖苷类、大环内酯类、四环素类及叶酸通道拮抗剂4类耐药基因。分为3个ST型,其中ST7型12株、ST1型5株、ST373型1株。全基因组序列分析显示2005年疫情菌株与四川省暴发疫情菌株同源,2006-2020年分离菌株基因组多态性大。结论江西省人感染猪链球菌病均为散发,菌株基因组多态性大,2005年疫情菌株与四川省暴发菌株相关,随后年份为多点平行传播。This study was aimed at understanding the pathogenic and genomic characteristics of Streptococcus suis strains in Jiangxi Province from 2005 to 2020,to provide a reference for the prevention and control of human infections with S.suis.We amplified 16S rRNA,cps2j,mr,sly,ef and gapdh virulence genes in the S.suis genome through polymerase chain reaction.The antibiotic sensitivity of the strains was determined according to the minimum inhibitory concentration.Genetic polymorphisms among strains were analyzed through whole genome sequencing.All ten strains of S.suis from humans and eight strains of S.suis from swine were of serotype 2.The virulence gene profiles differed,and at least three virulence genes were identified.The resistance rates to azithromycin and tetracycline were as high as 89.89%and 77.78%,respectively,and four types of resistance genes were carried:aminoglycosides,macrolides,tetracyclines,and folic acid channel antagonists.Three sequence types(STs)were identified:12 ST7,5 ST1,and 1 ST373.Whole genome sequencing indicated that the epidemic strains in 2005 showed homology with those in Sichuan Province,and the isolates from 2006 to 2020 had many genomic polymorphisms.S.suis infection in Jiangxi Province is sporadic,and the genomic polymorphism of the strains is high.The epidemic strains in 2005 were related to the outbreak strains in Sichuan Province,and multi-point parallel transmission was observed in subsequent years.
