摘 要:目的评价2种方法诱导实验性自身免疫性葡萄膜炎(EAU)青紫蓝兔模型的发生过程及特点。方法42只青紫蓝兔随机分为对照组(CG)6只、模型1组(MG1)18只、模型2组(MG2)18只。CG组不做任何处理;MG1组使用结核分枝杆菌H37Ra与弗氏完全佐剂的混悬液皮下注射进行预先免疫,19 d后右眼玻璃体腔注射结核分枝杆菌H37Ra建立EAU模型;MG2组采用牛血清白蛋白(BSA)耳缘静脉及玻璃体腔二次注射法建立EAU模型。所有动物分别于完成免疫后第1、4、7、14、21、28天行裂隙灯显微镜、前置镜、眼前节照相、眼B超、眼底照相观察眼部情况,根据自拟的EAU青紫蓝兔模型症状评分标准进行症状评分。结果(1)发病时间与周期:MG1组所有青紫蓝兔在完成诱导后第1天均初发炎症,第4天炎症达到高峰,第7天开始逐渐消退,症状持续4周以上。MG2组13只(72.22%)青紫蓝兔在完成诱导后第1天初发炎症,第4天炎症达到高峰,之后炎症开始逐渐消退,以眼前段炎症为主,症状持续4周以上;其余5只(27.78%)初发症状时间及症状高峰时间不一。(2)症状表现:2组青紫蓝兔眼部表现均为混合充血、角膜水肿、前房炎症及渗出、瞳孔受损、玻璃体混浊、视网膜出血。(3)症状评分:与CG组比较,MG1、MG2组青紫蓝兔各时间眼部症状评分均升高(MG1:t_(1d)=12.550,t_(4d)=12.620,t_(7d)=14.090,t_(14d)=11.610,t_(21d)=12.530,t_(28d)=41.470,均P=0.000。MG2:t_(1d)=2.600,P=0.016;t_(4d)=7.293,t_(7d)=8.354,t_(14d)=7.410,t_(21d)=6.843,t_(28d)=7.397,均P=0.000);与MG1组比较,MG2组青紫蓝兔诱导后第1、4天眼部症状评分降低(t_(1d)=4.062,P=0.000;t_(4d)=2.359,P=0.024),差异均有统计学意义。其余时间2组间眼部症状评分比较,差异均无统计学意义(P>0.05)。结论2种EAU青紫蓝兔模型其眼部表现与人类葡萄膜炎相似,MG1的初发炎症时间、炎症高峰时间、炎症严重程度及炎症变化趋势均一致。MG1较MG2的稳定性更好,可�OBJECTIVE To evaluate two methods for inducing experimental autoimmune uveitis(EAU)in chinchilla rabbit,including the onset process and characteristics of the disease.METHODS A total of 42 chinchilla rabbits were randomly divided into a control group(CG)of six rabbits,model 1 group(MG1)of 18 rabbits,and model 2 group(MG2)of 18 rabbits.The CG was not treated.The MG1 was pre-immunized with a suspension of H37Ra mycobacterium tuberculosis and Freund's complete adjuvant by subcutaneous injection,and EAU model was established by injecting mycobacterium tuberculosis H37Ra into the vitreous cavity of the right eye 19 days later.The MG2 was established by ear vein injection of bovine serum albumin(BSA)and vitreous cavity injection of BSA for the second time.All animals underwent slit-lamp microscopy,anterior segment photography,eye ultrasound,and fundus photography to observe eye conditions on days 1,4,7,14,21,and 28 after completing the immunization.Symptom scores were assessed based on a self-designed EAU chinchilla rabbit model symptom scoring system.RESULTS(1)Onset time and duration:All chinchilla rabbits in the MG1 developed inflammation on the first day after induction,peaked on the fourth day,gradually subsided on the seventh day,and lasted for more than four weeks.There were 13 rabbits(72.22%)in the MG2 developed inflammation on the first day after induction,peaked on the fourth day,and then gradually subsided.The inflammation was mainly in the anterior segment,and the symptoms lasted for more than four weeks;The onset time and peak time of the remaining five rabbits(27.78%)were different.(2)Symptoms:The eye manifestations of the two groups of chinchilla rabbits were mixed congestion,corneal edema,anterior chamber inflammation and exudation,pupil damage,vitreous opacity,and retinal hemorrhage.(3)Symptom scores:Compared with the CG,the eye symptom scores of chinchilla rabbits in the MG1 and MG2 increased at all time(MG1:t_(1d)=12.550,t_(4d)=12.620,t_(7d)=14.090,t_(14d)=11.610,t_(21d)=12.530,t_(28d)=41.470,all P=0.
