摘 要:目的 探索醒脾养儿颗粒对正常大鼠膀胱尿动力学及尿道外括约肌活动性的影响。方法 雄性SD大鼠60只,随机分为正常对照组、醒脾养儿颗粒9.2、4.6、2.3 g/kg组及奥昔布宁组,每组12只。各给药组灌胃给药,正常对照组给予等体积纯水,连续7 d,末次灌胃后分别于深麻醉和浅麻醉状态下测定大鼠排尿时膀胱内压和尿道外括约肌电活动。苏木素-伊红(HE)染色观察大鼠膀胱组织形态学变化。结果 醒脾养儿颗粒各剂量组能明显降低深麻醉及浅麻醉下大鼠基础膀胱压,醒脾养儿颗粒大剂量组能显著增加膀胱最大容量;醒脾养儿颗粒大剂量组明显增加浅麻醉下大鼠排尿P_(2)期波峰数及膀胱顺应性;各组膀胱组织形态未见明显差异。结论 醒脾养儿颗粒可显著降低基础膀胱压,增加膀胱最大容量和膀胱顺应性,促进膀胱持续有力的排尿,其作用可能与松弛膀胱平滑肌改善膀胱功能状态相关。Objective To investigate the effect of the Xingpi Yanger granules on bladder urodynamics and bladder external sphincter activity in normal rats.Methods Sixty healthy animals were divided into five groups:a control group;Xingpi Yanger granule 9.2,4.6,and 2.3 g/kg groups;and oxybutynin group,with 12 rats in each group.Experimental groups were given the drug by gavage for 7 consecutive days,and the control group was given equal volumes of pure water in the same way.After the last administration,we measured intravesical pressure and external urethral sphincter electromyography under light anesthesia and deep anesthesia when the rats were urinating.Rats’bladder pathomorphology was observed by hematoxylin-eosin staining.Results Xingpi Yanger granules obviously decreased rat basal bladder pressure(BBP)under light and deep anesthesia.The high-dose group,especially,showed significantly improved maximum bladder capacity,peak number of P_(2) phase,and bladder compliance under light anesthesia.There were no significant differences in the structural changes to the bladder in any group.Conclusions Xingpi Yanger granules significantly reduced BBP,increased the maximum bladder capacity and bladder compliance,and vigorously promoted bladder urination.The mechanisms may be related to the relaxation of bladder smooth muscle and improvements in bladder functional status.
