摘  要:茶树是重要的经济作物,生产上以扦插为主要的繁殖方式。在无性繁殖过程中,不定根的形成对茶树的成活至关重要。SBP(SQUAMOSA promoter-binding protein like)转录因子对侧生器官发生和不定根再生等植物生长发育过程起重要的调控作用。本研究通过生物信息学方法鉴定茶树SBP基因家族成员,以‘陕茶1号’为研究材料,克隆CsSBP24基因,检测其转录活性和亚细胞定位并在转基因烟草中对其进行功能分析。从茶树‘舒茶早’全基因组数据库中鉴定出24个SBP基因家族成员,分为6个亚家族。CsSBP24基因编码序列长度为930 bp,编码309个氨基酸;CsSBP24是定位于细胞核的转录因子。当过表达CsSBP24,不定根发生率、根粗和根长增加,但数量下降。结果表明,CsSBP24参与不定根发育过程的调控。本研究结果为后续利用分子手段调控茶树不定根发生提供了依据。Tea trees are important economic crops.The production is mainly based on cutting propagation.The formation of adventitious roots is crucial to the survival of tea trees asexual reproduction.SBP(SQUAMOSA promoter-binding protein like)transcription factor plays an important role in regulating the growth and development of plants such as lateral organogenesis and adventitious root regeneration.In this study,bioinformatics methods were used to identify the SBP gene family members in the tea plant.'Shaancha No.1'was used as the research material,the CsSBP24 gene was cloned,its transcriptional activity and subcellular location were detected,and the function of transgenic tobacco was analyzed.24 members of the SBP gene family were identified from the'Shuchazao'genome-wide database of tea trees,which were divided into 6 subfamilies.The length of CsSBP24gene coding sequence is 930 bp and encodes 309 amino acids;CsSBP24 is a transcription factor and located in the nucleus.When CsSBP24 was overexpressed,the incidence,root thickness and root length of adventitious roots were increased,but the number decreased.The result showed that,CsSBP24 participates in the adventitious roots regulation.Our results provide a basis for subsequent use molecular means to regulate the adventitious roots development of tea plant.
