摘  要:目的:探讨细胞连接相关基因在肾癌中的作用。方法:筛选细胞连接相关差异基因,Lasso-Cox回归分析构建风险模型,对风险模型进行生存分析及临床病理特征分析,UCSC验证风险模型用于临床的可行性;R包计算风险基因与免疫细胞的相关性;基于风险模型基因采用CellMiner数据库发现潜在的肾癌治疗药物,GSVA寻找风险评分差异通路。筛选高低风险组间差异核心基因,分析核心基因的表达在肾癌中的临床意义。初步探讨核心基因表达与风险评分高低及风险基因间的相关性。结果:基于细胞连接相关基因风险模型,高风险评分是肾癌患者生存预后的独立危险因素。白介素6(IL6)为高低风险组间核心差异基因,IL6高表达缩短高风险组肾癌生存预后而对低风险组肾癌患者生存预后无影响;IL6及肾癌临床特征构建的列线图可用于评估肾癌患者的生存预后;IL6与风险基因具有相关性。结论:IL6高表达缩短细胞连接相关基因介导的肾癌患者生存预后。Objective:To explore the significance of ellular contivity in renal cancer.Methods:A gene risk model was established for wide cell conectivity pathways by Lasso-Cox regression analysis and clinical survival data was downloaded from UCSC to verify the feasibility of risk score,and then the significance of risk model was explored in kidney cancer.Based on risk model genes,CllMiner database was used to discover potential kidney cancer therapeutics drugs.and GSVA chose differential pathways on base of risk score.Differential genes were screened between high and low risk groups and the core gene was determined.Next,we explored significance of the core gene in kidney cancer Results:Based on the gene risk model of extensive cell connectivity pathways,high risk score was an independent risk factor for the survival prognosis of renal cancer patients and was associated with immune cell infiltration.IL6 was the core differential gene between high and low risk groups,high exPression of IL6 shortened the survival prognosis of patients in.the high risk group,but had no effect on the surviwal of patients in the low risk group.The nomogram constructed by IL6 and clinical features has clinical value in the prognosis of kidney cancer.IL6 was associated with risk genes.Conclusion:The cll connection pathway risk gene model has important clinical application value in kidney cancer,and IL6 gene afected the survival prognosis of kidney cancer patients by interacting with risk genes.
