摘  要:以火龙果种子在无菌条件下萌发的幼苗作为外植体供体,子叶切段作为初代培养接种材料,通过调节不同外源激素及其浓度组合,选择最佳初代培养基、继代培养基和生根培养基,对建立火龙果组培育苗技术体系进行研究。结果表明:(1)种子经预处理后,用75%酒精浸泡10 s,再用0.1%Hg Cl_(2)浸泡5min的消毒效果最好,污染率为11.67%,萌发率为92.48%;(2)初代培养的最适培养基为MS+6-BA 2.0 mg·L^(-1)+NAA 0.2 mg·L^(-1)+蔗糖30 g·L^(-1),不定芽诱导率高达98.89%;(3)继代培养的最适培养基为MS+6-BA1.5 mg·L^(-1)+NAA 0.2 mg·L^(-1)+蔗糖30g·L^(-1),不定芽增殖系数达到6.52,不定芽粗壮,长势旺盛;(4)最佳生根培养基为1/2MS+NAA 0.5 mg·L^(-1)+IBA 0.1 mg·L^(-1)+蔗糖30 g·L^(-1)+AC 500 mg·L^(-1),生根率达98.89%,生根数为6.40条;(5)生根苗移栽30 d后成活率达97%。本研究以种子获取无菌苗作为外植体建立了火龙果组培技术体系,较茎段作为外植体有污染低,取材方便的优点。Taking pitaya seedlings sprouted under sterile conditions as explant donors,and cotyledon cuts as inoculation material of primary culture,the optimal primary culture medium,subculture culture medium and rooting medium were selected by adjusting combinations of different exogenous hormones and their concentrations,so as to study how to establish breeding technology system of pitaya tissue culture.The results showed that:(1)Immersing the seeds in 75%alcohol for 10 s and then in 0.1%HgCl_(2)for 5 min after pretreatment could gain optimal disinfection,with a contamination rate of 11.67%,and germination rate 92.48%;(2)the optimal medium for primary culture was MS+6-BA 2.0mg⋅L^(-1)+NAA 0.2mg⋅L^(-1)+sucrose 30g⋅L^(-1),leading to adventitious bud induction rate up to 98.89%;(3)MS+6-BA 1.5 mg⋅L^(-1)+NAA 0.2 mg⋅L^(-1)+sucrose 30g⋅L^(-1)was the most suitable for the subculture,leading to multiplication coefficient of 6.52,stout adventitious buds that grew vigorously;(4)the optimal rooting medium was 1/2 MS+NAA 0.5 mg⋅L^(-1)+IBA 0.1 mg⋅L^(-1)+sucrose 30g⋅L^(-1)+AC 500mg⋅L^(-1),leading to a rooting rate of 98.89%and rooting number 6.40;(5)survival rate of rooting plantlets reached 97%30d after transplantion.In this study,the tissue culture technology systemof pitayawas established by using sterile seedlings as explant donors,and compared with using stemsegments as explants,it has the advantages of lowpollution and convenient sampling.
