摘  要:以染化行业污染地块土壤中分散橙76作为研究对象,基于QSAR理论,运用T.E.S.T.软件预测分散橙76对大鼠的急性毒性LD50为3345 mg·kg^(-1).在此基础上设计开展大鼠经口毒性实验,结果表明,分散橙76主要对大鼠血液中红细胞相关指标产生显著影响,对体重增长、外观及内脏的影响变化无统计学意义.分散橙76未观察到有害作用的最低剂量值(NOAEL)为50 mg·kg^(-1)·d^(-1).以染毒后雌鼠的平均红细胞血红蛋白浓度(MCHC)作为代表性指标,运用基准剂量法(BMD),选择最优模型Hill模型拟合分散橙76的基准剂量下限值(BMDL)为20.38 mg·kg^(-1)·d^(-1),并通过不确定系数UF=10000推导得到人体经口非致癌毒性参数RfDo为0.002038 mg·kg^(-1)·d^(-1).最后以上海高黏性土壤为例,推导得到分散橙76在本土环境中一类用地和二类用地中的土壤限值分别为68.5 mg·kg^(-1)和537.3 mg·kg^(-1).分散橙76的毒性研究以及在土壤中标准限值的研究推导和确定目前在国内尚属首次,可为该类污染物的毒性评估和管控标准制订提供技术支撑.This research object is to establish a new modeling method of deriving the soil limit value for these unscaled contaminants in the dyeing and chemical industry lands.Take Disperse Orange 76 as an example(Abbreviated as“DO 76”below).Based on QSAR theory,the acute toxicity LD50 of DO 76 in rats,predicted by T.E.S.T.software,is 3345 mg·kg^(-1).Therefore,the toxicity experiment for NOAEL was designed and carried out.The experimental results showed that DO 76 mainly affected the red blood cell related indexes in the blood of the rats,and there was no statistical significance in the weight gain of the rats at each stage,the appearance and internal organs.The lowest no-observed adverse effect dose(NOAEL)of DO 76 was determined to be 50 mg·kg^(-1)·d^(-1) at this experimental dose.The optimal model-Hill model was used to calibrate the BMDL value at 20.38 mg·kg^(-1)·d^(-1).The reference dose,indicated as human oral non-carcinogenic toxicity parameter,was derived as 0.002038 mg·kg^(-1)·d^(-1) by the uncertainty coefficient 10000.In this study,the standard soil limits of DO 76 were derived to be 68.5 mg·kg^(-1) and 537.3 mg·kg^(-1) for the high cohensive soil in residential land and industrial land in urban area,respectively.The soil standard derivation of unscaled contaminants is an innovative trial to provide technical support for the toxicity assessment and control standards of this type of pollutants.
