异丹叶大黄素对肺腺癌A549细胞miR-654-3p表达及增殖、迁移、侵袭能力的影响

摘　　要：目的:探讨异丹叶大黄素(ISO)对肺腺癌A549细胞miR-654-3p表达及增殖、迁移、侵袭能力的影响。方法:用不同浓度(0、5、10、20μmol/L)ISO培养A549细胞24 h,采用CCK-8法检测细胞增殖抑制率,克隆形成实验检测克隆形成能力,划痕实验检测细胞迁移能力,Transwell小室检测细胞侵袭能力;采用实时荧光定量PCR法检测miR-654-3p表达水平,Western blot法检测E-cadherin、N-cadherin蛋白表达水平。取A549细胞,采用脂质体转染法分别转染miR-NC和miR-654-3p模拟物48 h,再加20μmol/L ISO孵育24 h,然后检测上述指标。另取A549细胞分别转染anti-miR-NC、anti-miR-654-3p,经20μmol/L ISO孵育后,检测上述指标。结果:ISO干预后,A549细胞中miR-654-3p表达量增加;细胞增殖抑制率升高,克隆形成数降低;侵袭细胞数、划痕愈合率、N-cadherin蛋白表达水平降低,E-cadherin蛋白表达水平升高;上述指标的变化均呈剂量依赖性(P<0.05)。与miR-NC组比较,miR-654-3p模拟物组克隆形成数减少,增殖抑制率升高(P<0.05);划痕愈合率、侵袭细胞数、N-cadherin蛋白表达水平降低(P<0.05),E-cadherin蛋白表达水平升高(P<0.05)。与anti-miR-NC组比较,anti-miR-654-3p组克隆形成数增高,增殖抑制率降低;划痕愈合率、侵袭细胞数、N-cadherin蛋白表达水平增高,E-cadherin蛋白表达水平降低(P<0.05)。结论:ISO可通过上调miR-654-3p表达抑制肺腺癌细胞的增殖、迁移及侵袭能力。Aim:To investigate the effects of isorhapontigenin(ISO)on miR-654-3p expression and proliferation,migration and invasion of lung adenocarcinoma A549 cells.Method:A549 cells were cultured by different concentrations(0,5,10 and 20μmol/L)ISO for 24 hours,and inhibition rate of cell proliferation was detected by CCK-8 assay,the ability of cloning,by cloning test,migration capacity,by scratch test,and invasion capacity,by Transwell test;the expression of miR-654-3p was detected by real-time-quantitative PCR,and E-cadherin,N-cadherin proteins expression levels were detected by Western blot.A549 cells were transfected with miR-NC or miR-654-3p mimic,or transfected with anti-miR-NC or anti-miR-654-3p,respectively;then cultured with 20μmol/L ISO for 24 hours.The indexes mentioned above were detected.Result:After being cultured with ISO,the expression of miR-654-3p,the inhibition rate of cell proliferation and E-cadherin protein expression of A549 cells increased,while the number of cell clones,invasive cells,the rate of scar healing,and N-cadherin protein expression decreased in a dose-dependent manner(P<0.05).Compared with miR-NC group,the inhibition rate of cell proliferation and E-cadherin protein expression of miR-654-3p mimic group were higher,while the number of cell clones,invasive cells,the rate of scar healing,and N-cadherin protein expression were lower(P<0.05).Compared with anti-miR-NC group,the inhibition rate of cell proliferation and E-cadherin protein expression of anti-miR-654-3p group were lower,while the number of cell clones,invasive cells,the rate of scar healing,and N-cadherin protein expression were higher(P<0.05).Conclusion:ISO could inhibit the proliferation,migration and invasion of A549 cells by up-regulating miR-654-3p.
