摘 要:目的:分析喉乳头状瘤(laryngeal papilloma,LP)中长链非编码RNA(long chain non-coding RNA,lncRNA)的差异表达谱并预测其靶基因,为喉乳头状瘤的治疗及预防提供新的思路及靶点。方法:用RNA-seq技术对喉乳头状瘤组织及瘤旁组织测序,以差异倍数(FC)作为标准筛选差异表达lncRNAs,分析并提取10 kb范围内lncRNA顺式调控靶标基因,并进入基因本体论(GO)和京都基因与基因组百科全书(KEGG)分析其生物学功能及参与信号通路等。结果:共筛选出227个差异表达基因(Log2FC≥2或≤0.5,P<0.05),其中高表达的上调基因76个(33%)、下调基因151个(67%)。满足位置要求和共表达关系的lncRNA顺式调控靶标基因共有44个,并进入GO和KEGG通路富集分析结果显示,差异表达基因GO BP显著富集在信号传导,KEGG通路显著富集在MAPK信号通路及PI3K-Akt信号通路。结论:lncRNA差异表达及其靶基因以及相关信号通路可能与喉乳头状瘤复发及其癌变密切相关。Objective:To analyze the differential expression profile of long chain non-coding RNA(lncRNA)in laryngeal papilloma(LP)and predict its target genes,and provide new ideas and targets for the treatment and prevention of laryngeal papilloma.Methods:The RNA sequencing(RNA-seq)technologies was used for the sequencing in laryngeal papilloma and adjacent tissue.The multiples differences fold change(FC)was as a standard for screening differentially expressed genes,and lncRNA cis-regulatory target genes in the range of 10 kb were analyzed and extracted,and their biological functions and participation in signaling pathways were analyzed through Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genome(KEGG),etc.Results:A total of 227 differentially expressed lncRNAs were selected(Log2FC≥2 or≤0.5,P<0.05),of which 76(33%)were highly expressed up-regulated genes and 151(67%)were down-regulated genes.There were 44 lncRNA cis-regulatory target genes that met the positional requirements and co-expression relationships,and the results of enrichment analysis into GO and KEGG pathways showed that differentially expressed genes GO BP were significantly enriched in signaling and KEGG pathway were significantly enriched in MAPK signaling pathway and PI3K-Akt signaling pathway.Conclusion:Differential expression of lncRNA and its target genes as well as related signaling pathways may be closely associated with laryngeal papilloma recurrence and its carcinogenesis.
