摘  要:目的探讨细胞因子信号转导抑制因子3(SOCS3)在慢加急性肝衰竭(ACLF)小鼠模型中的表达变化。方法24只小鼠随机分为ACLF组(A组,18只)、肝纤维化组(B组,3只)和正常对照组(C组,3只)。其中,B组腹腔注射四氯化碳5 mL/kg建立肝纤维化模型,A组腹腔注射四氯化碳5 mL/kg以及脂多糖20μg/kg、D-(+)-半乳糖胺盐酸盐2 g/kg建立ACLF模型。采用HE染色和Masson染色观察肝脏病理学变化,全自动生化分析仪检测血清ALT、AST和胆碱酯酶水平,实时荧光定量PCR检测肝脏组织中SOCS3、IL-6和IL-33 mRNA表达。结果HE染色和Masson染色证实A组小鼠肝脏组织符合ACLF病理学变化。与B组和C组相比,A组造模后8、16 h时ALT、AST水平升高(P<0.05),胆碱酯酶水平降低(P<0.05),且造模后16 h时变化更加明显(P<0.05)。与B组和C组相比,A组造模后4、8、16 h时SOCS3、IL-6、IL-33 mRNA表达均增加(P<0.05),造模后8、16 h时增加更加明显(P<0.05)。小鼠肝脏组织中SOCS3 mRNA表达与ALT、AST水平和IL-6、IL-33 mRNA表达均呈正相关(r=0.738、0.733和0.855、0.909,P<0.05),与胆碱酯酶水平呈负相关(r=-0.832,P<0.05)。结论ACLF小鼠肝脏组织中SOCS3 mRNA表达增加,并与生化指标和炎症细胞因子有关,提示SOCS3可能在ACLF炎症发生和发展中起重要作用。Objective To investigate the change of cytokine signal transduction inhibitor 3(SOCS3)expression in the rat model with acute-on-chronic liver failure(ACLF).Methods Twentyfour rats were randomly divided into three groups of A(18rats),B(3rats)and C(3rats).The rats in group B were intraperitoneally injected with carbon tetrachloride 5mL/kg for establishing a hepatic fibrosis model,while which in group A were intraperitoneally injected with carbon tetrachloride 5mL/kg,lipopolysaccharide 20μg/kg and D-gal 2 g/kg for establishing a ACLF model.The pathological change of liver tissues was observed by HE staining and Masson staining.The levels of serum ALT,AST and cholinesterase were measured by fully automatic biochemical analyzer.The mRNA expressions of SOCS3,IL-6and IL-33in the liver tissues were detected by real-time fluorescence quantification PCR.Results Both HE staining and Masson staining confirmed that the liver tissues of the rats in group A were in accord with the pathological change of ACLF.Compared with groups of B and C,the levels of ALT and AST were increased,while the level of cholinesterase was decreased at the 8th and 16th hour after model establishment in group A(P<0.05),which were changed more obviously at the 16th hour(P<0.05).Compared with groups of B and C,the mRNA expressions of SOCS3,IL-6and IL-33 were increased at the 4th,8th and 16th hour after model established in group A(P<0.05),which were increased more significantly at the 8th and 16th hour(P<0.05).The mRNA expression of SOCS3in the liver tissues of rats was positively correlated with the levels of ALT and AST and the mRNA expressions of IL-6and IL-33(r=0.738,0.733and 0.855,0.909,P<0.05),and was negatively correlated with the level of cholinesterase(r=-0.832,P<0.05).Conclusion The mRNA expression of SOCS3is obviously increased in the liver tissues of rats with ACLF,and is closely related with the biochemical criterion and inflammatory cytokines,which indicates that SOCS3may play an important role in the development and progression of ACLF.
