摘  要:在海洋微藻中,微拟球藻是最有潜力合成可食用二十碳五烯酸(EPA)的藻种之一.以富含EPA的微拟球藻磷脂为底物,筛选适合乙醇醇解催化EPA磷脂制备乙酯型EPA的脂肪酶,优化其催化工艺.实验研究表明,固定化酶Novozym 435是最适合催化微拟球藻磷脂制备EPA的生物催化剂.经优化催化条件(酶加量为磷脂量的15%,含水量为底物总量的2%,醇油质量比9∶1,反应温度40℃,反应时间12 h)后,Novozym 435催化磷脂的最高乙酯转化率为98.33%.结果表明,Novozym 435可能是潜在催化微拟球藻磷脂高值化生产开发EPA的脂肪酶.Among marine microalgae,Nannochloropsis is one of the most promising species for synthesizing edible eicosapentaenoic acid(EPA).A suitable lipase was screened to produce EPA as ethyl ester form by enzymatic ethanolysis of Nannochloropsis⁃derived phospholipids rich in EPA;meantime,the parameters of this process were optimized.The obtained results showed that the best biocatalyst for catalyzing the Nannochloropsis phospholipids was Novozym 435.Moreover,Novozym 435 gave the highest ethyl ester conversion rate of 98.33%under the optimized conditions(enzyme loading of 15%(based on phospholipids),water content of 2%(based on the total substrates),ethanol to phospholipids weight ratio of 9∶1,reaction temperature of 40℃,and reaction time of 12 h).Based on these obtained results,it can be concluded that Novozym 435 could be the poten⁃tial lipase for producing high⁃value EPA from Nannochloropsis phospholipids.
