摘 要:目的:探讨补肾活血汤调控PI3K/AKT/mTOR信号通路对绝经后骨质疏松症小鼠模型骨丢失的影响及其作用机制。方法:通过OVX法建立绝经后骨质疏松小鼠模型,将40只SD雌性小鼠随机分为假手术组、模型组、西药(戊酸雌二醇)组、补肾活血汤组,每组10只。除假手术组外,其他组均进行卵巢切除术构建骨质疏松小鼠模型,造模成功后,进行药物处理,假手术组、模型组给予等量的生理盐水处理,1次/d,持续6周。结果:与假手术组比较,模型组血清BALP水平降低、TRAP水平升高、骨密度降低,差异有统计学意义(P<0.05);与模型组相比较,干预组(西药组、补肾活血汤组)血清BALP水平升高、TRAP水平降低、骨密度升高,差异有统计学意义(P<0.05);与假手术组比较,模型组PI3K、AKT、mTOR表达降低,差异有统计学意义(P<0.05);与模型组相比较,干预组PI3K、AKT、mTOR表达升高,差异有统计学意义(P<0.05)。结论:补肾活血汤干预OVX小鼠骨质疏松模型,有减少骨丢失、抑制骨吸收的作用,其作用机制可能与PI3K/AKT/mTOR信号通路激活相关。Objective:To investigate the effect of Bushen Huoxue decoction on bone loss in a mouse model of postmenopausal osteoporosis by regulating PI3K/AKT/mTOR pathway and its mechanism.Methods:A mouse model of postmenopausal osteoporosis was established by OVX method.Forty SD female mice were randomLy divided into sham operation group,model group,western medicine(estradiol valerate)group and Bushen Huoxue decoction group.There were 10 rats in each group.Except for the sham operation group,the mice in the other groups were treated with ovariectomy to establish the osteoporosis model.After the model was successfully established,the mice in the sham operation group and the model group were treated with the same amount of normal saline once a day for 6 weeks.Results:Compared with the sham-operation group,the serum BALP level was decreased,the TRAP level was increased,and the BMD was decreased in the model group(P<0.05).Compared with the model group,the intervention group(western medicine group and Bushen Huoxue decoction group)had a significant increase in serum BALP level,a significant reduction in TRAP level,and a significant increase in BMD(P<0.05).Compared with the sham-operation group,the expression of PI3K,AKT and mTOR in the model group decreased,and the difference was statistically significant(P<0.05).Compared with the model group,the expression of PI3K,AKT and mTOR in the intervention group was increased,and the difference was statistically significant(P<0.05).Conclusion:Bushen Huoxue Decoction can reduce bone loss and inhibit bone resorption in OVX mice model of osteoporosis,which may be related to the activation of PI3K/AKT/mTOR signaling pathway.
