摘 要:金丝李(Garcinia paucinervis)是集材用、观赏、药用多种用途的珍贵树种,具有较高的经济价值。为揭示金丝李叶绿体基因组结构特点以及系统发育位置,本研究利用Illumina HiSeqTM4000平台对其叶绿体基因组进行测序,通过生物信息学方法分析其序列结构、基因特征、重复序列等,并构建系统发育树。结果表明,金丝李叶绿体基因组全长为157649 bp,含有85935 bp的大单拷贝(LSC)、17738 bp的小单拷贝(SSC)以及一对26988 bp反向重复(IRs)区。不同区域GC含量差异明显,大小依次为IR>LSC>SSC。该种共编码126个基因,包括81个编码蛋白基因(PCGs)、37个tRNA基因(tRNAs)和8个rRNA基因(rRNAs)。共鉴定出96个SSR,以单碱基重复为主,且偏好碱基A和T。金丝李IR边界无明显的扩张或收缩,但位于IR区边界的基因有差异;系统发育树显示金丝李与同属的莽吉柿(Garcinia mangostana)、大果藤黄(Garcinia pedunculata)、岭南山竹子(Garcinia oblongifolia)、棱果藤黄(Garcinia gummi-gutta)聚在同一分支上,且与岭南山竹子和棱果藤黄关系较近。本研究为金丝李的分子标记开发、种间遗传多样性以及系统发育研究提供一定的参考。Garcinia paucinervis is a rare tree species with higher economic value because of the timber,ornamental and medicinal utilization.In order to reveal the structural feature of chloroplast genome and its phylogenetic position,the Illumina HiSeqTM 4000 platform is used for sequence.Then the sequence structure,gene characteristics,repeat sequences and phylogenetic tree are further analyzed by bioinformatics methods.The results showed that the chloroplast genome was 157649 bp in length that contains large single copy(LSC)of 85935 bp,small single copy(SSC)of 17738 bp and a pair of inverted repeats(IRs)regions of 26988 bp.The GC content of different regions was significantly different,and the size order was IR>LSC>SSC.126 genes were identified,including 81 protein-coding genes(PCGs),37 tRNA genes(tRNAs)and 8 rRNA genes(rRNAs).96 SSR loci were detected,which were composed of single-base repeatswith nucleobase Aand T.There was no obvious expansion or contraction in the IR boundary,but the genes were different.Further,the phylogenetic tree showed G.paucinervis,G.mangostana,G.pedunculata,G.oblongifolia and G.gummi-gutta were clustered into the same branch.Moreover,G.paucinervis was more closely related to G.oblongifolia and G.gummi-gutta.This study will provide theoretical support for the development of analytical markers,interspecific genetic diversity,and phylogenetic analysis of this species.
