摘 要:目的探讨苏氨酰tRNA合成酶(TARS)在肝细胞癌(HCC)中的表达及其与患者预后、肝癌细胞增殖、迁移、侵袭的关系。方法收集93例HCC患者的临床病理资料,用免疫组织化学检测肝癌组织和癌旁组织中TARS的表达差异,分析其与患者临床病理特征及预后的关系;使用小干扰RNA(siRNA)技术下调肝癌细胞内TARS的表达,细胞计数试剂盒(CCK-8)实验检测细胞的增殖能力;Transwell小室检测细胞的迁移、侵袭能力,组间比较采用单因素和多因素方差分析及t检验。结果肝癌组织中TARS的表达明显高于癌旁组织(60.2%比25.8,P<0.01);TARS高表达组的整体生存期明显低于TARS低表达组(P<0.01),TARS高表达是影响HCC患者整体生存独立风险因素(P<0.01)。siRNA组细胞中TARS mRNA表达(0.04±0.01比0.99±0.03,P<0.01)及蛋白(0.18±0.02比1.11±0.07,P<0.01)表达低于对照组。CCK-8实验结果表明,与对照组比较,TARS降表达组细胞吸光度值在第3天(3.08±0.03比3.65±0.14,P<0.01)、第4天(3.94±0.06比5.42±0.15,P<0.01)、第5天(5.41±0.31比8.20±0.60,P<0.01)明显低于对照组,细胞增殖速度明显减慢;Transwell试验结果显示TARS降表达组细胞迁移能力明显低于对照组(66.8±4.7比77.8±7.2,P<0.01),TARS降表达组细胞侵袭数量低于对照组(37.0±4.9比49.2±5.0,P<0.01)。结论TARS在肝癌组织中异常高表达,为HCC患者预后的独立预测因素;敲低肝癌细胞中TARS的表达水平能够抑制肝癌细胞的增殖、迁移和侵袭。Objective To investigate the expression of threonyl-tRNA synthetase(TARS)in hepatocellular carcinoma(HCC)and the correlation with prognosis,cell proliferation,migration and invasion.Methods The clinical and pathological data of 93 patients with HCC were collected and the expression of TARS in HCC and adjacent tissues was detected by immunohistochemistry.The expression of TARS in HCC cells was down-regulated by small interfering RNA(siRNA)technology.Cell proliferation was detected by cell counting kit-8(CCK-8)assay and cell migration and invasion were measured by Transwell assays.Single and multiple factor ANOVA and t test were used in the experiment.Results The expression of TARS in hepatocellular carcinoma tissues was significantly higher than that in adjacent tissues(60.2%vs.25.8%,P<0.01);the overall survival of the high expression group of TARS were significantly worse than this of the low expression group of TARS(P<0.01),and high TARS expression was an independent risk factor for overall survival in HCC patients(P<0.01).The expression of TARS mRNA(0.04±0.01 vs.0.99±0.03,P<0.01)and protein(0.18±0.02 vs.1.11±0.07,P<0.01)in the TARS lowered expression group were lower than that in the control group.The results of CCK-8 experiments showed that the absorbance values of cells in the TARS lowered expression group were significantly decreased on day 3(3.08±0.03 vs.3.65±0.14,P<0.01),day 4(3.94±0.06 vs.5.42±0.15,P<0.01),and day 5(5.41±0.31 vs.8.20±0.60,P<0.01)than that in control group.The cell proliferation rate in the TARS lowered expression group was significantly reduced.Transwell assay showed that the migration ability in the TARS lowered expression group was decreased than that in control group[number of transmembrane cells:(66.8±4.7)cells vs.(77.8±7.2)cells,P<0.01],the invasion ability in the TARS lowered expression group was decreased than that in control group[number of transmembrane cells:(37.0±4.9)cells vs.(49.2±5.0)cells,P<0.01].Conclusion The expression of TARS is abnormally high in HCC,and t
