摘　　要：目的探讨Rab蛋白22A(Rab22A)在骨肉瘤组织中得表达及对肿瘤细胞增殖、迁移、侵袭和上皮-间充质转化的影响。方法选取2019年1月至2021年1月河南省人民医院收治的77例骨肉瘤组织和癌旁组织作为研究对象,采用蛋白质免疫印迹分析肿瘤组织和癌旁组织中Rab22A蛋白表达水平。采用对照慢病毒和Rab22A短发卡RNA(shRNA)慢病毒感染人骨肉瘤细胞U2OS,分别为对照组和Rab22A KD组,采用细胞计数试剂盒(CCK-8)分析两组细胞增殖能力;采用划痕实验、Transwell和蛋白质免疫印迹(Western blot)分析对照组和Rab22A KD组细胞迁移、侵袭和上皮-间充质转化能力。组间数据比较采用t检验。结果骨肉瘤组织中Rab22A表达水平(2.09±0.22)明显高于癌旁组织(1.01±0.18),差异有统计学意义(t=32.910,P<0.05)。对照组细胞吸光度(A)值(2.09±0.11)明显高于Rab22A KD组(1.53±0.11),差异有统计学意义(t=8.795,P<0.05)。对照组细胞细胞核增殖抗原(Ki-67)表达水平(1.18±0.12)明显高于Rab22A KD组(0.89±0.77),差异有统计学意义(t=5.195,P<0.05)。对照组细胞划痕愈合率[(85.98±3.82)%]明显高于Rab22A KD组[(68.03±5.64)%],差异有统计学意义(t=6.454,P<0.05)。对照组细胞侵袭数量[(117.67±12.23)个]明显高于Rab22A KD组[(87.83±5.88)个],差异有统计学意义(t=5.387,P<0.05)。对照组细胞黏着斑激酶(FAK)表达水平(0.98±0.12)明显高于Rab22A KD组(0.70±0.07),差异有统计学意义(t=5.039,P<0.05)。Rab22A对骨肉瘤细胞上皮-间充质转化的影响,Western blot结果显示,对照组细胞上皮细胞标志物E-钙黏蛋白(E-cadherin)表达水平(0.71±0.10)明显低于Rab22A KD组(1.11±0.09),差异有统计学意义(t=6.214,P<0.05)。对照组细胞间皮细胞标志物Snail、N-钙黏蛋白(N-cadherin)、波形蛋白(Vimentin)表达水平(0.98±0.10、1.16±0.09、1.09±0.11)明显低于Rab22A KD组(0.82±0.05、0.80±0.09、0.80±0.09),差异有统计学意义(t=3.595、6.849、4.931,P<0.05)。结Objective To investigate the expression of Rab protein 22A(Rab22A)in osteosarcoma and its effect on tumor cell migration,invasion and epithelial mesenchymal transformation.Methods A total of 77 cases of osteosarcoma and adjacent tissues treated in our hospital from January 2019 to January 2021 were selected as research objects,and protein immunoblotting was used to analyze the expression level of Rab22A protein in tumor tissues and adjacent tissues.Human osteosarcoma U2OS cells were infected with control lentivirus and Rab22A short hairpin RNA(shRNA)lentivirus,serving as the control group and Rab22A KD group,respectively.The cell proliferation ability of the two groups was analyzed by cell counting kit-8(CCK-8).The ability of cell migration,invasion and epithelial mesenchymal transformation in the control group and Rab22A KD group was tested by wounding healing,Transwell and Western blotting,respectively.T-test was used to compare the data between groups.Results The expression level of Rab22A in osteosarcoma(2.09±0.22)was significantly higher than that in adjacent tissues(1.01±0.18,t=32.910,P<0.05).The absorbance(A)value of control group(2.09±0.11)was significantly higher than that of Rab22A KD group(1.53±0.11,t=8.795,P<0.05).The expression level of proliferation cell nuclear antigen(Ki-67)in control group(1.18±0.12)was significantly higher than that in Rab22A KD group(0.890.77,t=5.195,P<0.05).The scratch healing rate of the control group[(85.98±3.82)%]was significantly higher than that of the Rab22A KD group[(68.03±5.64)%,t=6.454,P<0.05].The number of cell invasions in the control group(117.67±12.23)was significantly higher than that in the Rab22A KD group(87.83±5.88,t=5.387,P<0.05).The expression level of focal adhesion kinase(FAK)in control group(0.98±0.12)was significantly higher than that in Rab22A KD group(0.70±0.07,t=5.039,P<0.05).The expression level of E-cadherin in the control group(0.71±0.10)was significantly lower than that in the Rab22A KD group(1.11±0.09,t=6.214,P<0.05).The expression l
