摘  要:[目的]探讨miR-608调控胃癌细胞SGC-7901凋亡的潜在机制。[方法]将胃癌中表达失调的miRNA以mimics的形式在胃癌细胞SGC-7901中过表达,检测细胞的凋亡水平。将能够调节凋亡的miRNA在多种胃癌细胞中敲低或过表达,检测这些胃癌细胞的凋亡水平。miRDB和荧光素酶报告实验验证miRNA的潜在底物。胃癌细胞SGC-7901中过表达或敲低底物后,检测凋亡水平。[结果] miR-608能够抑制胃癌细胞SGC-7901的凋亡水平(26.31±6.54 vs 7.04±2.19,P<0.05)。在胃癌细胞株MKN-1、 MNK-28、MNK-45、HGC-27、SNU-1、SNU-5、 Hs-746T、SGC-7901、SNU-719、KATO 3中过表达miR-608后,这些胃癌细胞株的凋亡水平均显著下降(P<0.05)。胃癌细胞SGC-7901中过表达miR-608后,PCDHGA9的表达水平显著下降(1.42±0.31 vs 0.42±0.10,P<0.05)。此外,miR-608靶向PCDHGA9的3′端非编码区(100±7 vs 23±5,P<0.05)。敲低PCDHGA9后胃癌细胞SGC-7901的凋亡水平下降(20.04±2.00 vs 5.98±1.49,P<0.05)。过表达PCDHGA9后胃癌细胞SGC-7901的凋亡水平上升(21.42±4.03 vs 78.11±21.12,P<0.05)。[结论] miR-608通过靶向PCDHGA9的mRNA的3′端非编码区抑制PCDHGA9的翻译,从而抑制胃癌细胞SGC-7901的凋亡。[Objective]To investigate the potential mechanism of miR-608 in regulating the apoptosis of gastric cancer cells sgc-7901.[Method]The dysregulated miRNAs in gastric cancer were overexpressed in gastric cancer cells SGC-7901 in the form of mimics,and the apoptosis level of the cells was detected.miRNAs capable of regulating apoptosis were knocked down or overexpressed in various gastric cancer cells,and the apoptosis levels of these gastric cancer cells were detected.miRDB and luciferase reporter assays validate potential substrates for miRNAs.After overexpression or knockdown of substrates in gastric cancer cells SGC-7901,the level of apoptosis was detected.[Result]miR-608 could inhibit the apoptosis level of gastric cancer cell SGC-7901(26.31±6.54 vs 7.04±2.19,P<0.05).After overexpression of miR-608 in gastric cancer cell lines MKN-1,MNK-28,MNK-45,HGC-27,SNU-1,SNU-5,Hs-746T,SGC-7901,SNU-719,KATO 3,Apoptosis levels of these gastric cancer cell lines were significantly decreased(P<0.05).After overexpression of miR-608 in gastric cancer cell SGC-7901,the expression level of PCDHGA9 was significantly decreased(1.42±0.31 vs 0.42±0.10,P<0.05).Furthermore,miR-608 targeted the 3′ untranslated region of PCDHGA9(100±7 vs 23±5,P<0.05).The apoptosis level of gastric cancer cell SGC-7901 decreased after PCDHGA9 knockdown(20.04±2.00 vs 5.98±1.49,P<0.05).The apoptosis level of gastric cancer cell SGC-7901 increased after overexpression of PCDHGA9(21.42±4.03 vs 78.11±21.12,P<0.05).[Conclusion] miR-608 inhibited the translation of PCDHGA9 by targeting the 3′-UTR of PCDHGA9 mRNA,thereby inhibiting the apoptosis of gastric cancer cell SGC-7901.
