摘  要:目的:探讨长链非编码RNA(lncRNA)lnc AL137789.1对结直肠癌细胞增殖和迁移的影响。方法:选取2017年6月-2020年1月就诊于我院且就诊临床资料完整的40例结直肠癌患者,收集患者经手术切除的癌组织(结直肠癌组)及距癌组织>2 cm的癌旁组织(正常组),采用quantitative real-time PCR和Western blot检测结直肠癌组织和细胞中lnc AL137789.1和JAK2的表达水平。采用免疫组织化学(IHC)检测结直肠癌组织和邻近正常组织中JAK2的阳性率。采用EdU和Transwell实验检测结直肠癌细胞的增殖、迁移情况;采用皮尔逊相关系数分析lnc AL137789.1和JAK2的相关性。结果:结直肠癌细胞和组织中lnc AL137789.1的相对表达量明显高于癌旁正常组(P均<0.05),结直肠癌细胞和组织中JAK2的相对表达量明显高于癌旁正常组(P均<0.05)。与SH-lnc AL137789.1-NC组相比,SH-lnc AL137789.1显著降低了JAK2蛋白的表达(P均<0.05);与LV-lnc AL137789.1-NC组相比,LV-lnc AL137789.1显著提高了JAK2蛋白的表达(P均<0.05)。皮尔逊相关系数分析结果显示,JAK2和lnc AL137789.1呈正相关性(R^(2)=0.4015,P<0.0001)。与SH-lnc AL137789.1-NC组比,SH-lnc AL137789.1组显著抑制结直肠癌细胞的增殖、迁移(P均<0.05),而SH-lnc AL137789.1+JAK2组则显著促进结直肠癌细胞的增殖、迁移(P均<0.05)。与LV-lnc AL137789.1-NC组比,LV-lnc AL137789.1组显著促进结直肠癌细胞的增殖、迁移(P均<0.05),而LV-lnc AL137789.1+si-JAK2组则显著抑制结直肠癌细胞的增殖、迁移(P均<0.05)。结论:Lnc AL137789.1通过结构化修饰JAK2蛋白调节结直肠癌细胞增殖、迁移能力。Objective:To investigate the effect of long non-coding RNA(lncRNA)lnc AL137789.1 on the proliferation and migration of colorectal cancer cells.Method:A total of 40 patients with colorectal cancer who were admitted to Hainan Cancer Hospital from June 2017 to January 2020 and had complete clinical data were enrolled.Surgically resected cancer tissues(colorectal cancer group)and adjacent tissues>2 cm away from cancer tissues(normal group)were collected.Quantitative real-time PCR and Western blot were used to detect the expression levels of lnc AL137789.1 and JAK2 in colorectal cancer tissues and cells.The positive rate of JAK2 in colorectal cancer tissues and adjacent normal tissues was detected by immunohistochemistry(IHC).The proliferation and migration of colorectal cancer cells were detected by EdU and Transwell assay.The correlation between lnc AL137789.1 and JAK2 was analyzed by Pearson correlation coefficient.Results:The relative expression of lnc AL137789.1 in cells and tissues of colorectal cancer group was significantly higher than that in adjacent normal group(P<0.05),and the relative expression of JAK2 in cells and tissues of colorectal cancer group was significantly higher than that in normal group(P<0.05).Compared with SH-lnc AL137789.1-NC group,SH-lnc AL137789.1 group significantly decreased the expression of JAK2 protein(P<0.05).Compared with LV-lnc AL137789.1-NC group,LV-lnc AL137789.1 group significantly promoted the expression of JAK2 protein(P<0.05).Pearson correlation coefficient analysis showed that JAK2 was positively correlated with lnc AL137789.1 group(R^(2)=0.4015,P<0.0001).Compared with SH-lnc AL137789.1-NC group,SH-lnc AL137789.1 group significantly inhibited the proliferation and migration of colorectal cancer cells(P<0.05),while SH-LNC AL137789.1+JAK2 group significantly promoted the proliferation and migration of colorectal cancer cells(P<0.05).Compared with LV-lnc AL137789.1-NC group,LV-lnc AL137789.1 group significantly promoted the proliferation and migration of colorectal cancer ce
