摘 要:目的 探讨大蒜素对2型糖尿病(T2DM)大鼠肝脏组织病理学改变的影响。方法 取55只大鼠给予高脂高糖饮食6周后,采用腹腔注射链尿佐菌素的方法制备T2DM模型;另取10只大鼠作为正常对照组,给予常规饲料喂养。取50只造模成功大鼠随机分为模型组、二甲双胍组、低剂量大蒜素组、中剂量大蒜素组和高剂量大蒜素组。二甲双胍组大鼠灌胃给予二甲双胍300 mg·kg^(-1),每日1次,共4周;低、中、高剂量大蒜素组大鼠分别灌胃给予大蒜素5、10、20 mg·kg^(-1),每日1次,共4周;正常对照组和模型组大鼠灌胃给予生理盐水(10 mL·kg^(-1)),每日1次,共4周。干预4周后,经尾静脉采集各组大鼠空腹血1 mL,测定空腹血糖(FBG)水平;称量各组大鼠体质量并计算肝脏系数;经腹主动脉采血5 mL,应用全自动生物化学分析仪检测各组大鼠血清丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)、总胆红素(TBIL)水平;观察各组大鼠肝脏大体外观变化,苏木精-伊红染色观察各组大鼠肝脏组织病理学变化,并计算非酒精性肝脏损伤评分(NAS);Masson染色法观察肝组织纤维化状况,计算胶原容积分数(CVF);应用透射电子显微镜观察肝细胞超微结构变化。结果 模型组大鼠的FBG水平和肝脏系数显著高于正常对照组,体质量显著低于正常对照组(P<0.05)。二甲双胍组、中剂量大蒜素组、高剂量大蒜素组大鼠的FBG水平和肝脏系数显著低于模型组,体质量显著高于模型组(P<0.05);低剂量大蒜素组与模型组大鼠的FBG水平、体质量及肝脏系数比较差异无统计学意义(P>0.05)。低剂量大蒜素组大鼠的FBG水平和肝脏系数显著高于二甲双胍组,体质量显著低于二甲双胍组(P<0.05);中剂量大蒜素组大鼠的FBG水平显著高于二甲双胍组(P<0.05);中剂量大蒜素组与二甲双胍组大鼠的体质量和肝脏系数比较差异无统计学意义(P>0.05);高剂量大蒜素组与二甲双胍组大鼠�Objective To investigate the effect of Allicin on the pathological changes of liver in rats with type 2 diabetes mellitus(T2DM).Methods Fifty-five rats were taken and fed with high-fat and high-sugar diet for 6 weeks,then the rats were intraperitoneally injected with streptozotocin to prepare T2DM models.Other 10 rats were taken as the normal control group and fed with convertional forage.Fifty T2DM rat were taken and randomly divided into model group,metformin group,low-dose Allicin group,medium-dose Allicin group and high-dose Allicin group.The rats in metformin group were given metformin 300 mg·kg^(-1) by intragastric administration,once a day for 4 weeks;the rats in the low-,medium-,high-dose Allicin groups were given Allicin 5,10,20 mg·kg^(-1) by intragastric administration,respectively,once a day for 4 weeks;the rats in normal control group and model group were given normal saline(10 mL·kg^(-1))by intragastric administration,once a day for 4 weeks.After 4 weeks of intervention,1 mL fasting blood was collected from rats in each group through the tail vein to determine the level of fasting blood glucose(FBG);the body mass of rats in each group was measured and the liver coefficient was calculated;5 mL blood was collected through abdominal aorta,and the levels of aminotransferase(ALT),aspartate aminotransferase(AST)and total bilirubin(TBIL)in serum of rats in each group were detected by automatic biochemical analyzer.The liver was collected to observe the changes of gross appearance of rats in each group.The liver histopathological changes of rats in each group were observed by hematoxylin-eosin staining,and the nonalcoholic fatty liver disease activity score(NAS)was calculated.The liver tissue fibrosis of rats in each group was observed by Masson staining,and the collagen volume fraction(CVF)was calculated.The ultrastructure of liver cells was observed by transmission electron microscope.Results The FBG level and liver coefficient of rats in the model group were significantly higher than those in the norma
