纳米紫杉醇磁流体热化疗对肺癌细胞的抑制作用

摘  要:目的通过观察纳米紫杉醇磁流体热化疗对肺癌A549细胞的作用及对小鼠A549裸鼠移植瘤生长的影响,探讨纳米紫杉醇磁流体热化疗治疗肺癌的可行性。方法通过化学共沉淀法配制纳米紫杉醇磁流体;体外培养肺癌A549细胞,并分为对照组、磁流体热疗组、化疗组及磁热化疗组,观察经过分组处理后的人肺癌A549细胞的光密度值、杀伤率、生长曲线、细胞形态变化、细胞周期及凋亡情况。建立人肺癌A549裸鼠移植瘤模型,将实验裸鼠随机分为对照组、磁流体热疗组、化疗组以及磁热化疗组,观察四组治疗后肿瘤体积和质量的变化、各组瘤体的病理表现。结果细胞试验中,随着紫杉醇浓度的增加,磁流体热疗温度恒定,人肺癌A549细胞增殖抑制逐渐增强,其抑制作用更明显,活细胞数的光密度值明显下降,细胞凋亡率逐渐升高(P<0.05);免疫组化得出磁流体热疗组及磁热化疗组中细胞周期停滞于S期。动物试验中,磁流体热疗组、化疗组及磁热化疗组小鼠肿瘤质量和体积均受到抑制,其中磁热化疗组肿瘤生长抑制最明显(P<0.05);光镜和电镜下观察到肿瘤细胞发生凋亡并出现坏死样改变。结论纳米紫杉醇磁热化疗具有明显的抑制肿瘤生长作用,且较单用化疗或磁流体热疗抗肿瘤效果更好。Objective To investigate the feasibility of nano paclitaxel magnetic fluid thermochemotherapy for lung cancer by observing the effect of nano paclitaxel magnetic fluid thermochemotherapy on lung cancer A549 cells and the effect on the growth of transplanted A549 tumors in nude mice.Methods Nano paclitaxel magnetic fluid was prepared by chemical coprecipitation method.In vitro,the lung cancer A549 cells cultured and divided them into the control group,the magnetic fluid hyperthermia group,the chemotherapy group,and the magnetic thermal chemotherapy group.The optical density,killing rate,growth curve,cell morphology changes,cell cycle,and apoptosis of human lung cancer A549 cells after grouping treatment were observed.In addition,the human lung cancer A549 nude mouse transplantation tumor models were established and the experimental nude mice were randomly divided into the control group,the magnetic fluid hyperthermia group,the chemotherapy group,and the magnetic thermal chemotherapy group.The changes in tumor volume and mass after the treatment in the four groups were observed,as well as the pathological manifestations of the tumor bodies in each group.Results In cell experiments,as the concentration of paclitaxel increased,the temperature of magnetic fluid hyperthermia remains constant,and the proliferation inhibition of human lung cancer A549 cells gradually increased(P<0.05).At the same time,the optical density value of the number of living cells significantly decreased,and the apoptosis rate gradually increased.Immunohistochemistry showed that the cell cycle in the magnetic fluid hyperthermia group and magnetic hyperthermia chemotherapy group stagnated in the S phase.In animal experiments,the tumor mass and volume of mice in the magnetic fluid hyperthermia group,chemotherapy group,and magnetic thermal chemotherapy group were all inhibited(P<0.05),with the magnetic thermal chemotherapy group showing the most significant tumor growth inhibition.The changes of apoptosis and necrotic were observed in tumor cells u
