摘　　要：目的探讨丝氨酸羟甲基转移酶2(SHMT2)在胃癌组织中的表达及其与临床病理特征和预后之间的关系,为胃癌诊断和预后预测提供新的生物标志物。方法使用免疫组化法检测SHMT2在胃癌、癌旁正常组织中的表达,并通过GEPIA数据库进行验证。利用胃癌组织芯片评估SHMT2与临床病理特征之间的关系。Kaplan-Meier生存分析和生信数据库比较和验证SHMT2高表达组和低表达组患者的生存差异,Cox回归分析不同因素对患者预后的影响。结果SHMT2在胃癌组织和癌旁正常组织中高表达率分别为65.96%和48.84%,差异有统计学意义(χ^(2)=5.396,P=0.020)。SHMT2表达水平与淋巴结转移(χ^(2)=12.214,P<0.001)、血管神经侵犯(χ^(2)=3.884,P=0.049)、临床分期(χ^(2)=13.455,P<0.001)有关。SHMT2高表达和低表达组生存曲线差异有统计学意义(χ^(2)=8.248,P=0.004),SHMT2高表达组总生存期较低表达组短。Cox回归分析显示淋巴结转移和血管神经侵犯是患者预后的独立危险因素(HR=3.704,P=0.002;HR=1.865,P=0.021)。结论SHMT2在胃癌组织中高表达,且与患者较差的总生存期相关,对判断患者预后具有一定意义。Objective To investigate the expression of serine hydroxymethyltransferase 2(SHMT2)in the gastric cancer and its relationship with clinical data and prognosis,so as to provide a mew biomarker for the diagnosis and prognosis of gastric cancer.Methods The expression of SHMT2 in the gastric cancer and adjacent normal tissues was detected by immunohistochemistry,and was verified by GEPIA database.The relationship between SHMT2 and clinicopathological characteristics was evaluated by using gastric cancer tissue microarray.Kaplan-Meier survival analysis and bioinformatics database was used to compare and verify the survival difference between patients in the high expression of SHMT2 group and the low expression of SHMT2 group,COX regression was used to analyze the impact of different factors on the prognosis of patients.Results The high expression rate of SHMT2 in the gastric cancer tissues and adjacent normal tissues was 65.96%and 48.84%,respectively,and the difference was statistical significance(χ^(2)=5.396,P=0.020).The expression level of SHMT2 was correlated with lymph node metastasis(χ^(2)=12.214,P<0.001),vascular and neural invasion(χ^(2)=3.884,P=0.049)and TNM stage(χ^(2)=13.455,P<0.001).Survival analysis showed that there was a significant difference in the survival curve between the high expression of SHMT2 group and the low expression of SHMT2 group(χ^(2)=8.248,P=0.004),the overall survival time of the high expression of SHMT2 group was shorter than that of the low expression of SHMT2 group.COX regression analysis showed that lymph node metastasis and vascular and nerve invasion were the risk factors for prognosis(HR=3.704,P=0.002;HR=1.865,P=0.021).Conclusion SHMT2 is highly expressed in the gastric cancer,and is associated with poor overall survival of patients,which has certain significance in judging the prognosis of patients.
