摘 要:目的探讨N-乙酰半胱氨酸对前列腺癌的作用及相关机制。方法体外培养人前列腺癌PC3细胞,用不同浓度N-乙酰半胱氨酸处理细胞,MTT和细胞克隆形成实验检测细胞活力,细胞划痕实验检测细胞迁移能力,流式细胞技术检测细胞凋亡,蛋白印迹检测细胞中p38,caspase3和MMP2的蛋白表达;体内建立荷瘤裸鼠模型,腹腔注射给予不同剂量N-乙酰半胱氨酸,检测肿瘤块体积和重量变化,蛋白印迹和RT-PCR法检测肿瘤组织中p38,caspase3和MMP2的表达。结果与对照组相比,N-乙酰半胱氨酸呈剂量依赖性抑制PC3细胞增殖,促进细胞凋亡和抑制细胞迁移能力(P<0.05),下调PC3细胞中p38表达水平,并上调caspase-3和MMP2表达水平(P<0.05);与对照组相比,N-乙酰半胱氨酸显著抑制荷瘤裸鼠肿瘤块的形成,并下调肿瘤组织中p38表达水平,上调caspase-3和MMP2表达水平(P<0.05)。结论N-乙酰半胱氨酸对前列腺癌具有潜在的治疗价值,并且其与抑制PC3细胞增殖和诱导细胞凋亡有关。Objective To observe the effect of N-acetylcysteine on prostate cancer and its related mechanisms.Methods Human prostate cancer pc3 cells were cultured in vitro,and the cells were treated with different concentrations of N-acetylcysteine.MTT and cell clone formation experiments were used to detect cell viability.Cell metastasis ability was tested using cell scratching assay and apoptosis was detected by flow cytometry.The protein expression levels of p38,caspase3 and MMP2 were detected by Western blot.A nude mouse model bearing tumor was established in vivo,and different doses of N-acetylcysteine were given intraperitoneally.Changes in tumor mass and weight were measured.Western blot and RT-PCR were used to detect the expression levels of p38,caspase3 and MMP2 in tumor tissues.Results Compared with the control group,N-acetylcysteine inhibited PC3 cell proliferation,promoted apoptosis and inhibited cell migration ability in a dose-dependent manner(P<0.05),and down-regulated p38 protein expression,as well as up-regulated the expression of caspase-3 and MMP2 protein in PC3 cells(P<0.05).Compared with the control group,N-acetylcysteine could significantly inhibit the formation of tumor masses in nude mice bearing tumors,and down-regulate p38 expression levels,and simultaneously up-regulate caspase-3 and MMP2 expression in tumor tissues(P<0.05).Conclusion N-acetylcysteine has potential therapeutic value for prostate cancer,and it is related to inhibiting cell proliferation and promoting apoptosis.
