摘 要:目的探讨miR-153-3p调控鼻咽癌细胞增殖、凋亡和周期的作用。方法利用生物信息学软件对miR-153-3p的候选靶基因进行预测。实时荧光定量PCR和Western blotting分别检测miR-153-3p对细胞内mRNA和蛋白表达的影响。用密理博MUSETM细胞检测仪对细胞周期和细胞凋亡情况进行检测。用CCK8法和克隆技术形成实验检测细胞增殖能力。结果生物信息学预测结果显示,miR-153-3p与BCL2分子之间存在一个高可信度的结合区。miR-153-3p mimic能上调miR-153-3p的表达,降低BCL2蛋白的表达。与对照组相比,过表达miR-153-3p的6-10B细胞的凋亡明显增加,增殖能力受到显著抑制。结论miR-153-3p通过下调BCL2的表达,能够促进鼻咽癌细胞凋亡,抑制其增殖。Objective To investigate the effect of miR-153-3p on proliferation,apoptosis and cycle of human nasopharyngeal carcinoma cells.Methods The candidate target genes of miR-153-3p were predicted by bioinformatics software.The effects of miR-153-3p on the expression of mRNA and protein in cells were detected by real-time fluorescence quantitative PCR and Western Blotting respectively.The cell cycle and apoptosis were detected by MILLIPORE MUSE cell detector.The ability of cell proliferation was detected by CCK8 and colony formation assay.Results The results of software prediction showed that there was a highly reliable binding region between miR-153-3p and BCL2 molecules.MiR-153-3p mimic up-regulated the expression of miR-153-3p and decrease the expression of BCL2 protein.Compared with the control group,the apoptosis of 6-10B cells overexpressing miR-153-3p was significantly increased,and the ability of proliferation was significantly inhibited.Conclusion miR-153-3p can promote apoptosis and inhibit proliferation of nasopharyngeal carcinoma cells by down-regulating the expression of BCL2.
