摘  要:为了研究禽类颗粒细胞的功能,建立一套良好的颗粒细胞培养体系至关重要。该研究分离培养了产蛋高峰期母鸡的6~8 mm小黄卵泡颗粒细胞,在细胞基质、不同物种血清、血清浓度上进行优化。结果表明,鼠尾胶原比明胶预铺更能促进颗粒细胞生长,细胞传代至4代才发生死亡。2.5%鸡血清与2.5%FBS(fetal calf serum)、2.5%鸡血清+2.5%FBS相比,2.5%鸡血清传代后细胞形态优于其他组。显微镜观察和EdU(5-Ethynyl-2’-deoxyuridine)检测表明,随着鸡血清浓度的增加,颗粒细胞增殖增加。该研究建立了在预铺鼠尾胶原作为胞外基质,采用含5%鸡血清的培养体系,使得鸡小黄卵泡颗粒细胞传代后仍能保证其增殖特性。In order to study the function of avian granulosa cells,it is important to establish a suitable granulosa cells culture system.In this study,granulosa cells of 6-8 mm small yellow follicle of hens were isolated and cultured,and cell matrix,serum of different species,and serum concentration in cells culture system were optimized.The results showed that rat tail collagen was better in promoting the growth of granulosa cells than gelatin pre-plating,and the cells did not die until passage 4.The cell morphology of 2.5% chicken serum was better than that of other groups.Microscopic observation and EdU detection showed that with the increase of serum concentration,granulosa cell proliferation increased.In this study,a pre-plated rat tail collagen was used as an extracellular matrix,and a culture system containing 5% chicken serum was used to ensure that the small yellow follicle granule cells of the chicken can be passaged to ensure its proliferation characteristics.
