小鼠老化对卵母细胞内钙库钙含量的影响

摘　　要：本研究以青、老年小鼠MⅡ卵母细胞为研究对象,首先利用Mito-tracker Red作为分子探针对青、老年卵母细胞线粒体的空间定位进比较。结果表明,老年小鼠线粒体定位异常的卵母细胞率明显高于青年小鼠。利用线粒体钙和内质网钙检测试剂盒分别对青、老年小鼠卵母细胞内线粒体钙和内质网钙含量进行测定。结果表明,老年小鼠卵母细胞内线粒体钙含量显著高于青年小鼠(P<0.05),老年小鼠卵母细胞内质网钙含量显著低于青年小鼠(P<0.05)。In this study,MⅡoocytes from young and old mice were used as the study subjects,and the spatial localization of mitochondria in young and old oocytes was firstly compared using Mito-tracker Red as a molecular probe.The results showed that the rate of oocytes with abnormal mitochondrial localization was significantly higher in aged mice than in young mice.Then,the mitochondrial calcium and endoplasmic reticulum calcium contents in oocytes of young and old mice were measured using mitochondrial calcium and endoplasmic reticulum calcium assay kits,respectively.The results showed that the mitochondrial calcium content in the oocytes of aged mice was significantly higher than that of young mice(P<0.05),the endoplasmic reticulum calcium content in the oocytes of aged mice was significantly lower than that of young mice(P<0.05).
