摘 要:[目的]从鹅粪便中分离筛选具有生产潜能的酵母菌,为水质净化、畜禽粪便的资源化利用、动物养殖等提供菌种资源。[方法]使用麦芽汁培养基,采用涂布法和平板划线方法从鹅粪便中分离酵母菌,然后对纯化的菌种进行形态学、生理生化鉴定和26S rDNA序列的同源性分析进行鉴定。同时测定酵母菌对模拟污水COD的降解性能。[结果]分离到4株酵母菌,通过鉴定和系统发育分析,最终将分离到的菌株Y-1、Y-2、Y-3、Y-5分别鉴定并命名为阿氏丝孢酵母菌Y-1(Trichosporon asahii Y-1)、红酵母菌Y-2(Rhodotorula sp.Y-2)、胶红酵母菌Y-3(Rhodotorula mucilaginosa Y-3)、隐球酵母菌Y-5(Cryptococcus albidus Y-5)。4株酵母菌中,Y-3对模拟污水COD的去除效果最好,在菌浓度为1.5×10^(9) CFU/mL、1%接种量、培养96 h时,对污水中COD的去除效果达到56.8%。[结论]筛选到4株酵母菌,其中胶红酵母菌Y-3对模拟污水COD的去除效果最好,可进一步用于后续研究。[Objective]To isolate and screen yeast with production potential from goose feces,and provide bacterial resources for water purification,resource utilization of livestock and poultry feces and animal breeding.[Method]Yeasts were isolated from goose feces using the wort medium by the plate coating method and the streak plate method,and according to morphological,physiological and biochemical,26S rDNA sequence analysis.At the same time,it was measured that the degradation performance of the yeast for COD in simulated sewage.[Result]The four isolated yeast strains Y-1,Y-2,Y-3 and Y-5 were finally named as Trichosporon asahii Y-1,Rhodotorula sp.Y-2,Rhodotorula mucilaginosa Y-3,Cryptococcus albidus Y-5 through identification and phylogenetic analysis.Among the 4 strains of yeast,Rhodotorula mucilaginosa Y-3 had the best removal effect on COD in simulated sewage,and the removal rate reached 56.8%when the bacterial concentration was 1.5×10^(9) CFU/mL,1%inoculum and cultured for 96 h.[Conclusion]Four strains of yeasts were screened,among which Rhodotorula mucilaginosa Y-3 had the best removal effect on COD in simulated sewage,which could be further used in subsequent studies.
