摘 要:虾青素(Astaxanthin,AST)是一种天然的脂溶性类胡萝卜素,已被证实有抗氧化、抗炎等多种生物活性,但AST对溃疡性结肠炎(Ulcerative colitis,UC)的影响尚不清楚。实验探究AST对葡聚糖硫酸钠(Dextran sulfate sodium,DSS)诱导的小鼠溃疡性结肠炎的潜在治疗作用。40只C57BL/6小鼠分为正常对照(Control)、模型组(Model)、溶剂对照橄榄油组(Oil)和虾青素(AST)4组。除Control组外,其他小鼠构建实验性慢性结肠炎模型。在造模的第5 d开始分别口服生理盐水、橄榄油或AST至观测末期。检测各组小鼠体重、粪便隐血和存活率等相关疾病表征;检测结肠长度及脾脏指数;H&E染色检测结肠病理情况;髓过氧化物酶(Myeloperoxidase,MPO)活性检测结肠组织中性粒细胞浸润情况;ELISA检测结肠与血清中炎性因子TNF-α和IL-1β水平。结果表明,AST治疗能缓解DSS诱导结肠炎小鼠的体重减轻(P<0.05)、结肠缩短(P<0.05)和疾病活动指数(Disease activity index,DAI)增加(P<0.01)。小鼠给予AST后,与对照组相比结肠长度增加(P<0.05),脾脏指数明显降低(P<0.01),并且由DSS诱导的结肠组织杯状细胞丢失,隐窝结构破环及大量炎性细胞浸润的病理情况得到改善(P<0.05)。同时,AST可降低结肠组织MPO酶活(P<0.05),降低结肠组织与血清中炎性因子TNF-α、IL-1β的含量(P<0.05)。实验证明AST可显著缓解DSS诱导的溃疡性结肠炎的疾病表征,改善结肠病理情况,降低炎性因子水平,为其开发用于治疗UC提供了依据。Astaxanthin(AST)is a natural fat-soluble carotenoid that has been shown to have a variety of biological activities including the antioxidant and anti-inflammatory,while the effect of AST on ulcerative colitis(UC)is unclear.The potential therapeutic effect of AST on dextran sulphate sodium(DSS)induced ulcerative colitis in mice was investigated.40 male C57BL/6 were divided into four groups:normal control(Control),Model(Model),olive oil(Oil)and astaxanthin(AST).Except for the Control group,other groups of mice were constructed as experimental chronic colitis models.Starting on day 5 of modeling,each group of mice was administered orally saline,olive oil or AST respectively until the end of observation.Relevant disease characteristics including body weight,fecal occult blood and survival rate were measured.Colon length and spleen index were investigated.Hematoxylin-eosin(H&E)staining was performed to detect colonic pathology.Myeloperoxidase(MPO)activity was used to detect neutrophil infiltration in colonic tissue.Enzyme-linked immuno sorbent assay(ELISA)was used to detect TNF-αand IL-1βlevels of inflammatory factors in colon and serum.The results showed that AST treatment alleviated the weight loss(P<0.05)and increased in Disease Activitv Index(DAI,P<0.01)in DSS-induced colitis mice.The mice were administered AST with increased colonic length(P<0.05),significantly reduced splenic index(P<0.01)compared to the Control group and improved DSS-induced pathological condition of colonic tissue with loss of goblet cells,destruction of crypt structures,and massive inflammatory cell infiltration(P<0.05).Moreover,AST treatment decreased MPO enzyme activity in the colonic tissue(P<0.05)and reduced the levels of inflammatory factors TNF-αand IL-1βin the colon and serum(P<0.05).Experiment demonstrated that AST significantly alleviated the disease characteristics,improved colonic pathology,reduced inflammatory factor levels of DSS-induced ulcerative colitis and suggested that AST supplement might be effective for the treatment
