摘 要:视网膜退行性疾病是全球范围内导致视力损害的重要因素,与其相关的研究日益成为热点。Sigma-1受体是一种由223个氨基酸构成的内质网跨膜蛋白,其基因序列在整个哺乳动物中高度保守,并广泛分布于各种组织和器官中。大量的研究证明了Sigma-1受体在视网膜病理生理过程中发挥保护作用,包括抗氧化应激、抗炎症反应、抗细胞凋亡等。此外,Sigma-1受体在视网膜疾病,特别是糖尿病视网膜病变、视网膜色素变性和青光眼视神经损伤中具有潜在的治疗作用。本文就Sigma-1受体介导的视网膜神经保护的分子机制及其在视网膜疾病治疗中的应用进行综述。Retinal degenerative diseases are a major contributor to visual impairment worldwide,and research related to retinal degenerative diseases is increasingly becoming a hot topic.The sigma-1 receptor is a 223-amino-acid endoplasmic reticulum transmembrane protein,and its gene sequence is highly conserved throughout mammals.Sigma-1 receptors are widely distributed in various tissues and organs.Numerous studies have proved the protective role of sigma-1 receptors in retinal pathological processes,including anti-oxidative stress,anti-inflammatory response,and anti-apoptosis.In addition,sigma-1 receptors have potential therapeutic roles in retinal diseases,especially diabetic retinopathy,retinitis pigmentosa and glaucomatous neuropathy.In this paper,the molecular mechanisms of sigma-1 receptor-mediated retinal neuroprotection and its application in the treatment of retinal diseases were reviewed.
