摘 要:为探究谷胱甘肽(glutathione,GSH)在17℃负压环境下对猪精液保存质量及抗氧化能力的影响,试验采集4头2~3岁的健康长白猪精液,取合格精液混合后分为4份,分别用未添加和添加1,5,10 mmol/L GSH的Modena稀释液稀释,均在17℃、-0.04 MPa条件下保存11 d,分别在保存前(第0天)和保存第1,3,5,7,9,11天检测精子活率、精子活力、精子畸形率及运动参数(平均路径速度、曲线线速度、鞭打频率)指标和精液的总抗氧化能力(T-AOC)及H_(2)O_(2)浓度。结果表明:第1,3,5,7,9,11天时,添加1,5,10 mmol/L GSH的精子活率、精子活力及运动参数[平均路径速度、曲线线速度、鞭打频率(除第1天外)]指标和精液的T-AOC均高于未添加GSH,精子畸形率、H_(2)O_(2)浓度(除第1天外)均低于未添加GSH。其中第11天时,添加5 mmol/L GSH的精子活率可达18.5%,显著高于添加1,10mmol/L GSH(P<0.05)。第7天时,添加5 mmol/L GSH的精子活力显著高于添加1,10 mmol/L GSH(P<0.05);第11天时,添加5 mmol/L GSH的精子活力显著高于添加1 mmol/L GSH(P<0.05),与10 mmol/L GSH差异不显著(P>0.05)。第7天时,添加5 mmol/L GSH的精子畸形率显著低于添加1,10 mmol/L GSH(P<0.05);第11天时,添加5 mmol/L GSH的精子畸形率最低,显著低于添加1 mmol/L GSH(P<0.05),但与添加10 mmol/L GSH差异不显著(P>0.05)。第11天时,添加5 mmol/L GSH的鞭打频率显著高于未添加和添加1,10 mmol/L GSH(P<0.05)。第9天时,添加1,5 mmol/L GSH的T-AOC显著高于添加10 mmol/L GSH(P<0.05)。第5,9,11天时,添加1,5 mmol/L GSH的H_(2)O_(2)浓度均显著低于添加10 mmol/L GSH(P<0.05)。说明在-0.04 MPa条件下添加GSH可提高17℃保存的猪精液的精子品质和抗氧化性能,其中添加5 mmol/L GSH最合适。To study the effects of glutathione(GSH)on the preservation quality and antioxidant capacity of boar semen under 17℃negative pressure,semen was collected from four healthy long white boars aged 2 to 3 years,and the qualified semen was mixed and divided into four parts,which were diluted with Modena dilution without adding or adding 1,5,10 mmol/L GSH concentration.The four parts of semen were all stored under 17℃and-0.04 MPa for 11 d.The viability,motility,malformation rate and motility parameters(average path velocity,curve line velocity,beat cross frequency)indexes of sperm and the antioxidant capacity(T-AOC)and H_(2)O_(2)concentration of semen were measured on days 0,1,3,5,7,9 and 11,respectively.The results showed that the viability,motility and motility parameters(average path velocity,curve line velocity,beat cross frequency)of sperm and the antioxidant capacity(T-AOC)of semen in the 1,5 and 10 mmol/L GSH groups were higher than those in the group without GSH on days 1,3,5,7,9 and 11,and the sperm malformation rate and H_(2)O_(2)concentration(except on day 1)were lower than those in the group without GSH.On the 11th day,the sperm viability of sperm in the 5 mmol/L GSH group was 18.5%,which was significantly higher than that in the 1 and 10 mmol/L GSH groups(P<0.05).On day 7,the sperm viability in the 5 mmol/L GSH group was significantly higher than that in the 1 and 10 mmol/L GSH groups(P<0.05).On day 11,the sperm motility in the 5 mmol/L GSH group was significantly higher than that in the 1 mmol/L GSH group(P<0.05),but had no significant difference compared with 10 mmol/L GSH group(P>0.05).On day 7,the sperm deformation rate in the 5 mmol/L GSH group was significantly lower than that in the 1 and 10 mmol/L GSH groups(P<0.05).On day 11,the sperm deformation rate in the 5 mmol/L GSH group was the lowest,which was significantly lower than that in the 1 mmol/L GSH group(P<0.05),but had no significant difference compared with 10 mmol/L GSH group(P<0.05).On day 11,the beat cross frequency in the 5 mmol/L GSH
