摘  要:为了从氧化应激角度探究阿魏酸对小鼠镉中毒致急性肾脏损伤的保护作用,试验建立氯化镉所致小鼠急性肾脏损伤模型,将80只小鼠随机分为空白组、模型组和阿魏酸低(75 mg/kg)、中(150 mg/kg)、高(300 mg/kg)剂量组,每组16只,空白组、模型组灌胃生理盐水,其余组连续灌胃不同剂量的阿魏酸,除空白组灌胃生理盐水外,其余组均在第15天灌胃氯化镉溶液(按体重0.2 mL/10 g),试验结束后,小鼠摘眼球取血,分离血清,检测血液生理指标[白细胞(WBC)、红细胞(RBC)、血红蛋白(HGB)和血细胞比容(HCT)]、血液生化指标[肌酐(Cr)、血尿素氮(BUN)和尿酸(UA)]。小鼠采血后采取颈椎脱臼法处死,采集肾脏组织,用苏木精-伊红(H.E.)染色后,用医学图像分析系统观察肾脏病理组织学变化;取肾脏组织制成10%和20%的组织匀浆,测定肾脏组织抗氧化指标[还原性谷胱甘肽(GSH)、超氧化物歧化酶(SOD)、丙二醛(MDA)和总抗氧化能力(T-AOC)]。结果表明:与模型组相比,阿魏酸各剂量组血液中WBC、RBC数量及HGB含量、HCT均升高,其中阿魏酸高剂量组WBC数量显著升高(P<0.05);阿魏酸低、中、高剂量组RBC数量显著升高(P<0.05)。与空白组相比,模型组Cr、BUN、UA浓度显著升高(P<0.05);与模型组相比,阿魏酸低、中、高剂量组Cr、BUN、UA浓度均显著降低(P<0.05);MDA浓度显著升高(P<0.05),GSH、SOD活性及T-AOC显著下降(P<0.05);阿魏酸中、高剂量组肾脏MDA浓度显著降低(P<0.05)。阿魏酸高剂量组肾脏组织病变明显减轻,效果好于阿魏酸低、中剂量组。说明高剂量阿魏酸对镉中毒导致的小鼠肾脏损伤具有一定的保护作用。In order to explore the protective effects of ferulic acid on acute kidney injury caused by cadmium poisoning in mice from the perspective of oxidative stress,in this experiment,a model of acute kidney injury caused by cadmium chloride was established.Eighty mice were randomly divided into blank group,model group,low(75 mg/kg),medium(150 mg/kg),and high(300 mg/kg)dose groups of ferulic acid.There were 16 animals in each group,and the blank group and model group had gavage with normal saline,and the rest groups had continuous gavage with ferulic acid.Except for the blank group by gavage with saline,the rest of the groups had gavage with cadmium chloride solution on the 15th day(0.2 mL/10 g by body weight).After the experiment,blood was taken from mouse eyeballs,and serum was isolated;blood physiological indexes[white blood cells(WBC),red blood cells(RBC),hemoglobin(HGB)and hematocrit(HCT)],blood biochemical indexes[creatinine(Cr),blood urea nitrogen(BUN)and uric acid(UA)]were determined.After blood collection,mice were sacrificed by cervical dislocation and kidney tissue was collected.After staining with hematoxylin-eosin(H.E.),the histopathological changes of the kidney were observed with a medical image analysis system.Kidney tissue was prepared into 10%and 20%homogenates to determine renal tissue antioxidant indexes[reducing glutathione(GSH),superoxide dismutase(SOD),malondialdehyde(MDA),and total antioxidant capacity(T-AOC)]were determined.The results showed that compared with the model group,the number of WBC,RBC and HGB,HCT were increased in the blood of all dose groups of ferulic acid,among which WBC was significantly increased in the ferulic acid high-dose group(P<0.05),and RBC in the low,medium and high dose groups of ferulic acid was significantly increased(P<0.05).Compared with the blank group,the concentrations of Cr,BUN and UA in the model group increased significantly(P<0.05).Compared with the model group,the concentrations of Cr,BUN and UA in the low,medium and high dose groups of ferulic acid decre
