摘  要:目的:探讨磷脂酰肌醇-3激酶调节亚单位1(PIK3R1)在人原发性肝细胞癌(HCC)中的表达水平及其与恶性肿瘤上皮间质转化的相关性。方法:选取80例HCC病人癌组织及相应癌旁组织,通过免疫组织化学染色法检测HCC组织及相应癌旁组织中PIK3R1编码蛋白(PIK3R1/p85α)表达,并检测HCC组织中EMT相关蛋白转化生长因子β(TGF-β)、上皮性钙黏蛋白(E-cadherin)表达,评估其表达与HCC病人临床特征的相关性。结果:PIK3R1/p85α在HCC组织中的阳性率为70.0%(56/80),高于癌旁组织的17.5%(7/40)(P<0.05);不同组织分化程度HCC病人的PIK3R1/p85α、E-cadherin和TGF-β表达差异均有统计学意义(P<0.05~P<0.01);HCC组织中PIK3R1/p85α与E-cadherin表达呈明显负相关关系(r=-0.323,P<0.01),与TGF-β表达呈正相关关系(r=0.247,P<0.05);无侵袭转移HCC病人和有侵袭转移病人的PIK3R1/p85α蛋白表达差异有统计学意义(P<0.05)。结论:PIK3R1在HCC组织中表达水平升高,高水平PIK3R1可能会促进肿瘤上皮间质转化的发生,并与HCC不良预后有关。Objective:To investigate the expression level of phosphatidylinositol-3 kinase regulatory subunit 1(PIK3R1)in human primary hepatocellular carcinoma(HCC)and its correlation with epithelial mesenchymal transformation of malignant tumor.Methods:The expression levels of PIK3R1 coding protein(PIK3R1/p85α)in HCC tissues and corresponding adjacent tissues of 80 HCC patients were detected by immunohistochemical staining.The expression levels of EMT-related proteins transforming growth factorβ(TGF-β)and epithelial cadherin(E-cadherin)in HCC tissue were detected,and the correlation of the expression with clinical characteristics of HCC patients was evaluated.Results:The positive rate of PIK3R1 expression in HCC tissue(70.0%)was significantly higher than that in corresponding adjacent tissue(17.5%)(P<0.05).The differences of the expression of PIK3R1/p85α,E-cadherin and TGF-βin HCC patients with different levels of tissue differentiation were statistically significant(P<0.05 to P<0.01).The expression of PIK3R1/p85αwas negatively correlated with E-cadherin expression(r=-0.323,P<0.01),and positively correlated with TGF-βexpression in HCC tissue(r=0.247,P<0.05).The difference of the protein expression of PIK3R1/p85αbetween non-invasive metastatic and invasive metastatic HCC patients was statistically significant(P<0.05).Conclusions:The expression level of PIK3R1 in HCC tissue increases.The high level of PIK3R1 may promote the epithelial mesenchymal transformation of tumors,and be associated with the poor prognosis of HCC.
