摘 要:山西省大同云州区是我国黄花菜的主要生产基地,为了探清大同市云州区黄花菜产区的主要品种,为大同市区域优良种质鉴定及选育提供理论依据,也为云州区黄花菜产区的品牌保护提供有力支持,试验采集大同市云州区33个村落的黄花菜种苗,采用形态学指标与山西农业大学黄花菜种质资源创新与利用课题组前期开发的43对高多态性EST-SSR分子标记对这些种苗进行遗传多样性分析、遗传结构分析与聚类分析。结果表明,根据花蕾性状的形态学指标进行聚类分析,可将大同地区黄花菜资源分为3个组群;筛选出18对EST-SSR标记能够很好地区分待测种质,18对EST-SSR标记对146份萱草属植物种质共扩增出88条多态片段,平均每对引物扩增出4.89条多态片段,引物多态性信息含量的平均值为0.2982,大同市大部分黄花菜种质资源与参考种质大同黄花的遗传相似系数均为1.0000。基于EST-SSR标记的聚类分析与遗传结构分析结果相差不大,能很好地对黄花菜种质进行分类,除了2个来自峰峪村和2个来自贺店村的黄花菜种质外,其余134份来自大同市云州区的黄花菜种质资源与大同黄花聚为一类。以上结果表明,除了来自峰峪村和贺店村的4个黄花菜种质外,其余134份来自大同市云州区的黄花菜种质资源全部为大同黄花。Yunzhou district,Datong,Shanxi province is the main production base of Hemerocallis citrina in China.In order to find out the main varieties of Hemerocallis citrina in Yunzhou district of Datong city and provide a theoretic basis for identification and breeding for the regional excellent varieties in Datong city,provide strong support for brand protection of the Hemerocallis citrina production area in Yunzhou district,in this experiment,Hemerocallis citrina seedlings from 33 villages in Yunzhou district of Datong city were collected,and morphological index and 43 pairs of highly polymorphic EST-SSR molecular markers previously developed by the research group on innovation and utilization of Hemerocallis citrina germplasm resources of Shanxi Agricultural University was used to conduct the genetic diversity analysis,genetic structure analysis,and cluster analysis of these seedlings.The results showed that the Hemerocallis citrina resources in Datong area could be divided into three groups by cluster analysis based on morphological index of flower bud traits.18 pairs of EST-SSR markers were screened out to be able to distinguish the tested germplasm.A total of 88 polymorphic fragments were amplified from 146 Hemerocallis citrina germplasm by these 18 pairs of EST-SSR markers,with an average of 4.89 polymorphic fragments amplified by each pair of primers.The average value of polymorphism information content(PIC)of primers was 0.2982.The genetic similarity coefficient between most of the Hemerocallis citrina germplasm resources in Datong and the reference germplasm Datong Huanghua was 1.0000.The results of cluster analysis and genetic structure analysis based on EST-SSR markers were similar,which could well classify the Hemerocallis citrina germplasm.Except for two Hemerocallis citrina germplasm from Fengyu village and two germplasm from Hedian village,the other 134 Hemerocallis citrina germplasm resources from Yunzhou district of Datong city were clustered with Datong Huanghua.The above results showed that except for
