摘 要:肺癌的发病率和死亡率居全球癌症第1位,在我国的发病率也居首位,而肺癌发生转移是治疗失败的主要原因,如何阻断肺癌转移已成为亟需解决的问题。现已发现诸多信号通路与肿瘤的发生、发展、转移有着密切关系。本研究总结其中主要的信号通路TGF-β/Smad、PI3K/AKT/mTOR、JAK/STAT、Wnt/β-catenin与肺癌转移的最新研究进展。以“肺癌、信号通路”为关键词,检索2018-01-01-2022-12-31中国知网及PubMed相关文献。纳入标准:肺癌和TGF-β/Smad信号通路的研究;肺癌和PI3K/AKT/mTOR信号通路的研究;肺癌和JAK/STAT信号通路的研究;肺癌和Wnt/β-catenin信号通路的研究。排除标准:肺癌和其他信号通路的相关研究。最终纳入分析文献51篇。结果表明,肺癌的发生和诸多信号通路相关,其中与TGF-β/Smad、PI3K/AKT/mTOR、JAK/STAT、Wnt/β-catenin信号通路密切相关。TGF-β/Smad、PI3K/AKT/mTOR、JAK/STAT、Wnt/β-catenin信号通路广泛参与了肺癌肿瘤细胞的生长、分化、侵袭、转移等多种过程。进一步研究与肺癌转移的相关通路,明确其各自的作用机制,有望发现更多有效的转移过程中的靶点。The incidence and mortality of lung cancer rank first worldwide and also rank first in incidence in our country,while lung cancer metastasis is a major cause of treatment failure,and how to block lung cancer metastasis has become a much-needed problem.Now,many signaling pathways have been found to be closely related to tumor development and metastasis.This review summarized the recent research progress on several major signaling pathways,including TGF-p/Smad,PI3K/Akt/mTOR,JAK/stat,and Wnt/β-catenin,and lung cancer metastasis.Using"lung cancer,signaling pathway"as keywords,we searched relevant literature in China National Knowledge Infrastructure and PubMed from January 1,2018 to December 31,2022.Inclusion criteria:studies of lung cancer and TGF-p/Smad signaling pathways;studies on lung cancer and the PI3K/Akt/mTOR signaling pathway;studies of lung cancer and JAK/STAT signaling pathways;lung cancer and Wnt/β-catenin signaling pathway.Exclusion criteria:relevant studies of lung cancer and other signaling pathways.Fifty-one articles were finally included in the analysis.The results showed that the carcinogenesis of lung cancer and many signaling pathways were related,among which,TGF-β/Smad signaling pathway,PI3K/Akt/mTOR signaling pathway,JAK/STAT signaling pathway,Wnt/β-catenin signaling pathway were closely related.TGF-β/Smad signaling pathway,PI3K/Akt/mTOR signaling pathway,JAK/STAT signaling pathway,Wnt/β-catenin signaling pathway were widely involved in many processes including lung cancer tumor cell growth,differentiation,invasion,metastasis.Further research on the related pathways with lung cancer metastasis,and clarifying their respective mechanisms of action will hopefully lead to the discovery of more effective targets in the metastatic process.
