摘  要:目的揭示2022年2月广西德保县本土新冠疫情中病例新型冠状病毒(SARS-CoV-2)基因组特征及变异情况,为广西新型冠状病毒肺炎(COVID-19)疫情溯源及防控工作提供科学依据。方法从广西德保县2月本土新冠疫情的确诊病例中选取30份病例的上呼吸道标本进行核酸提取、构建DNA测序文库、第二代测序仪上机测序、拼接测序数据,通过基因比对和构建进化树完成病毒基因变异位点和进化分析。结果与武汉参考株相比,广西德保县2月本土聚集性新冠疫情病例的SARS-CoV-2核苷酸突变中位数为57个(56~58个),氨基酸突变的中位数为63个(62~65个),同源性非常高,均属于VOC/Omicron变异株BA.1进化分支。结论SARS-CoV-2全基因组测序分析对新冠疫情的溯源研判提供科学依据,对新冠疫情的防控至关重要。Objective To disclose the genome characteristics and mutations of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)strains from COVID-19 outbreak in Guangxi Debao,and provide scientific reference for the prevention and control of COVID-19 in Guangxi.Method The viral nucleic of 30 specimens were extracted;the sequencing library was constructed following the manufacturing instructions,and the sequencing data were analyzed by bioinformatics software.Results Compared with the Wuhan reference strain,it was found that the median(minimum to maximum)number of the nucleotide mutations of the SARS-CoV-2 genome sequence of Guangxi Debao epidemic was 57(56-58)and the median(minimum to maximum)number of amino acid mutations was 63(62-65);all the SARS-CoV-2 of 30 cases were belong to VOC/Omicron mutant BA.1.Conclusion Continuous analysis of the sequences would be vital to tracing COVID-19 epidemic,which was vital to prevent and control of COVID-19 epidemic.
