摘 要:为探究[鱼安][鱼康]鱼皮酶溶胶原蛋白肽(pepsin-solubilized collagen peptides from Lophius litulo skin, PSCP)对高脂饮食诱导的小鼠非酒精性脂肪肝的影响及其机制,48只C57BL/6小鼠喂食高脂饲料4周诱导非酒精性脂肪肝模型,采用二甲双胍,低、中、高剂量PSCP处理模型小鼠6周,分别测定小鼠血清生理生化指标、肝脏氧化应激水平、肝脏组织病理学变化和蛋白的表达水平。实验结果表明:与模型组相比,二甲双胍和高剂量PSCP显著降低了小鼠血清中总胆固醇(total cholesterol, TC)、甘油三酯(triglyceride, TG)、低密度脂蛋白胆固醇(low-density lipoprotein, LDL-C)水平及谷草转氨酶(aspartate aminotransferase, AST)和谷丙转氨酶(alanime aminotransferase, ALT)活力,显著提高了高密度脂蛋白胆固醇(HDL-C)水平;二甲双胍及中、高剂量PSCP显著升高了小鼠肝脏中谷胱甘肽(glutathione, GSH)、超氧化物歧化酶(superoxide dismutase, SOD)、过氧化氢酶(catalase, CAT)含量,显著降低肝脏丙二醛(malondialdehyde, MDA)水平。苏木精-伊红染色和透射电镜结果显示,二甲双胍组和PSCP组小鼠肝脏结构明显好转。蛋白免疫印迹结果显示,与正常组相比,模型组小鼠肝脏中AMPKα、p-AMPKα和IκBα的蛋白表达量显著降低,SREBP-1和p65、p50、IKKα的蛋白表达量显著升高;而给予二甲双胍和PSCP后AMPKα、p-AMPKα和IκBα的蛋白表达量增加,SREBP-1和p65、p50、IKKα的蛋白表达量降低。研究结果表明,PSCP可显著改善非酒精性脂肪肝小鼠的生化指标,恢复肝组织结构,其改善机制可能与AMPK/NF-κB信号通路转导相关。To investigate the effect and mechanism of pepsin-solubilized collagen peptides from of Lophius litulo skin(PSCP)on high fat diet(HFD)induced non-alcoholic fatty liver disease(NAFLD)in C57BL/6 mice.Forty-eight C57BL/6 mice were fed with high-fat diet for 4 weeks to induce the nonalcoholic fatty liver disease model.The mice were then treated with metformin,low,medium and high dose PSCP for 6 weeks.The serum physiological and biochemical indexes,liver oxidative stress level,liver histopathology changes and protein expression levels of the mice in each group were measured respectively.The experimental results show that compared with the model group,total cholesterol(TC),triglyceride(TG),low-density lipoprotein(LDL-C),aspartate aminotransferase(AST)and alanine aminotransferase(ALT)levels in the positive drug group and high dose PSCP group were significantly decreased(P<0.05),and HDL-C levels were significantly increased(P<0.05).glutathione(GSH),superoxide Dismutase(SOD),and catalase(CAT)activities in liver of the positive drug group,middle dose and high dose PSCP groups were significantly increased(P<0.05),while malondialdehyde(MDA)contents were significantly decreased(P<0.05).Hematoxylin-eosin(HE)staining and transmission electron microscopy showed that the liver structure of the positive drug group and PSCP groups was significantly improved.The results of protein immunoblotting showed that,compared with the normal group,the protein expression levels of AMPKα,p-AMPKα,and IκBαin the liver of model group were significantly decreased(P<0.05),while the protein expression levels of SREBP-1,p65,p50,and IKKαwere significantly increased(P<0.05).However,after administration of the positive drug and PSCP,the protein expression levels of AMPKα,p-AMPKα,and IκBαwere increased,and the protein expression levels of SREBP-1,p65,p50,and IKKαwere decreased.The result showed that PSCP could effectively improve the biochemical indices of NAFLD mice and restore the liver tissue structure,and the mechanism might be related to AM
