摘 要:目的:探究Beclin-1和LC3在根尖肉芽肿、根尖囊肿和牙发育性囊肿中的表达和功能。方法:知情同意后收集检测18位患者牙源性病损组织,包括根尖肉芽肿(n=7)、根尖囊肿(n=8)和牙发育性囊肿(n=3)。通过免疫组化染色评估LC3和Beclin-1的表达和亚细胞定位,并通过实时CPR分析比较自噬(Beclin-1和LC3)与促炎因子基因表达的相关性。结果:Beclin-1和LC3蛋白在根尖肉芽肿和根尖囊肿中表达明显增加,但在牙发育性囊肿中表达少。在根尖肉芽肿中,阳性染色多分布在炎症细胞(巨噬细胞和淋巴细胞,以及多形核中性粒细胞),而在根尖囊肿中,阳性染色分布在炎症细胞、血管周围细胞和上皮细胞中。实时PCR结果显示BECLIN1和LC3B基因表达在根尖肉芽肿中最高,在牙发育性囊肿中最低。在根尖肉芽肿和根尖囊肿中,BECLIN1和IL-8之间以及LC3B和IL-1β、IL-8、IL-17之间有明显正相关性。结论:自噬可能在炎症性根尖周病变的免疫反应中起到保护作用。Objective:To investigate the expression pattern of Beclin-1 and LC3 and explore their possible function in apical granulomas,radicular cysts and dentigerous cysts.Methods:Eighteen human odontogenic lesions,including apical granulomas(n=7),radicular cysts(n=8)and dentigerous cysts(n=3),were collected and examined after informed consent.The expression and subcellular localization of LC3 and Beclin-1 in odontogenic lesions were evaluated by immunohistochemical staining.Their expression levels were correlated with conventional pro-inflammatroy cytokines by RT-PCR examinations.Results:Beclin-1 and LC3 protein significantly increased in apical granulomas and radicular cysts,but extremely decreased in dentigerous cysts.In apical granulomas,the positive staining was strongly expressed in inflammatory cells(macrophages and lymphocytes,and polymorphonuclear neutrophils),whereas in radicular cysts,positive staining was observed in inflammatory cells,perivascular cells and epithelial cells.Quantitative analysis revealed highest mRNA levels of BECLIN1 and LC3B in apical granulomas and lowest in dentigerous cysts.In both apical granulomas and radicular cysts,a strong significant positive correlations were observed between BECLIN1 and IL-8,and between LC3B and IL-1β,IL-8,IL-17.Conclusion:Autophagy may act as a protective function in immune response of inflammatory periapical lesions and consequently in radicular cysts development and growth.
