摘  要:在后基因组时代,天然产物挖掘因其巨大的应用前景一直备受生物医药领域的研究人员关注。随着研究者对天然产物的生物合成研究的不断深入,从海量的基因组数据中挖掘生物合成基因簇,并从中筛选新天然产物的研究策略,已经越来越受到人们的青睐。在众多的天然产物挖掘方法中,以自抗性基因为导向的挖掘,提供了一种新的分析天然产物合成途径的视角和一种高效的挖掘策略,筛选到的天然产物通常也具有较高的生物活性。重点介绍了天然产物的4种典型的自抗性机制,介绍了现有的抗性基因数据库,举例阐述了近几年出现的自抗性基因导向的天然产物挖掘策略,对基于计算机技术的微生物源天然产物的挖掘进行了展望。In the post-genomic era,discovery of natural product has been receiving great attention from researchers in the field of biomedicine due to its huge application prospects.With the continuous research on the biosynthesis of natural products,the research strategy of discovering of biosynthetic gene clusters from massive genomic data and screening new natural products has become more and more popular.Compared with other discovery methods of natural products,those guided by self-resistance genes provides a new perspective on the synthesis pathway of the natural product synthesis as well as an efficient strategy due to the high biological activity of the products.This review summarized four typical self-resistance mechanisms of natural products.The existing resistance gene databases were also reviewed.The newly emerged self-resistance gene guided discovery of natural products was illustrated with examples.Finally,future prospects for the discovery of microbial natural products based on the developed computer technology were also discussed.
