摘  要:松花菜是目前中国花椰菜消费的主要类型,松花性状是松花菜重要的经济性状。为了鉴别松花和紧花型花椰菜在分子水平上的差异,本研究以紧花型‘花椰菜F90’和松花型‘花椰菜S120’及其杂交F2分离群体为材料,从F2群体中挑选松花和紧花极端性状单株各40株构建DNA混池,以甘蓝HDEM为参考基因组,通过对松紧花DNA混池进行BSA重测序及Δ(SNP-index)分析,在花椰菜的C1染色体上获得一个长度为5.5 Mb的候选区间。根据该区间的插入/缺失(InDel)变异位点,开发InDel分子标记,经过筛选获得2个与花椰菜松花性状紧密连锁的InDel标记,在松紧花性状上存在显著差异。研究结果在一定程度上反映了松花和紧花性状在In Del标记上的差异,这些研究为精细定位松花性状候选基因以及分子标记辅助选育花椰菜松花新材料提供新思路。Loose-curd cauliflower is the main consumption characters of cauliflower in China,and the loose-curd is an important economic trait of cauliflower.In order to identify the genetic mechanism differences between the cauliflower of loose-curd and compact-curd.In this study,the compact-curd'cauliflowerF9o'and loose-curd'cauliflowerS120'and their hybrid F2 segregated populations were used as materials.40 loose-curd and 40 compact-curd individuals with extreme traits from a F2 segregating population were selected and grouped as two DNA mixed bulks,termed the loose-curd pool and compact-curd pool.Clean reads were aligned to the reference genome of Brassica oleracea L.HDEM.By BSA re-sequencing and analysis of the loose-curd and compact-curd of DNA mixed bulks,a candidate region with a length of 5.5 Mb was identified on chromosome C1 of cauliflower.According to the insertion/deletion(InDel)variable sites,InDel molecular markers were developed.After screening,two InDel markers closely linked to loose-curd traits of cauliflower were obtained and there were significant differences in the loose-curd and compact-curd traits.To a certain extent,the results reflect the differences between loose-curd and compact-curd traits in InDel markers.These studies provide new ideas for fine-mapping of candidate genes for loose-curd trait and molecular marker-assisted selection of new loose-curd cauliflower materials.
