摘  要:目的:探讨微管蛋白γ1(Tubulin gamma 1, TUBG1)在肝细胞肝癌(Liver hepatocellular carcinoma, LIHC)中的表达及其预后价值。方法:通过TIMER 2.0数据库对TUBG1基因进行泛癌分析;利用GEPIA和UALCAN数据库分析TUBG1基因在正常肝组织和肝细胞肝癌中的差异表达;使用UALCAN分析TUBG1基因与临床病理特征的相关性;利用GEPIA数据库分析TUBG1基因与临床预后的关系。最后,通过STRING数据库构建相互作用蛋白网络图并利用DAVID数据库进行功能富集分析。结果:TUBG1基因在多个癌症中均有差异表达,且TUBG1基因在LIHC组织中表达水平高于正常组织(P<0.05);TUBG1基因的表达量与肿瘤分期、患者种族、性别、年龄、体重、肿瘤分级等均紧密相关(P<0.05);在LIHC患者中,高表达的TUBG1总生存率和无病生存率均低于低表达者(P<0.05);与TUBG1关系紧密的20个蛋白,通过功能富集分析发现其编码基因共同参与了10个细胞组分、2个分子功能和9个生物学过程。结论:TUBG1基因在LIHC中的差异表达,与临床病理特征和预后不良均相关,有望成为LIHC诊断和预后的重要指标。Objective:To investigate tubulinγexpression of Tubulin gamma 1(TUBG1)in hepatocellular carcinoma(LIHC)and its prognostic value.Methods:Pan-cancer analysis of TUBG1 gene was made through TIMER 2.0 database;GEPIA and UALCAN databases were used to analyze the differential expression of TUBG1 gene in normal liver tissue and hepatocellular carcinoma;The correlation between the TUBG1 gene and clinicopathological features was analyzed by UALCAN.GEPIA database was used to analyze the relationship between TUBG1 gene and clinical prognosis.Finally,the network diagram of interacting proteins was constructed through String database,and the DAVID database was used for functional enrichment analysis.Results:TUBG1 gene was differentially expressed in multiple cancers,and the expression level of TUBG1 gene in hepatocellular carcinoma was higher than that in normal tissues(P<0.05);The expression of TUBG1 gene was closely related to tumor stage,patient race,gender,age,weight and tumor grade(P<0.05);In patients with hepatocellular carcinoma,the overall survival rate and disease-free survival rate of TUBG1 with high expression were lower than those with low expression(P<0.05).Through functional enrichment analysis,it was found that the coding genes of 20 proteins closely related to TUBG1 were involved in 10 cell components,2 molecular functions and 9 biological processes.Conclusion:The differential expression of TUBG1 gene in hepatocellular carcinoma is associated with clinicopathological features and poor prognosis.It is expected to become an important index of diagnosis and prognosis.
