摘 要:【目的】研究二氧化硫(sulfur dioxide, SO2)对木纳格葡萄果实中糖含量及糖代谢途径的影响。【方法】基于高效液相色谱法(High Performance Liquid Chromatography, HPLC)测定木纳格葡萄果实贮藏过程中蔗糖、葡萄糖和果糖含量变化,分析20个与糖代谢有关基因的表达差异性。【结果】SO2处理使得贮藏0~10 d时葡萄糖含量显著提高。贮藏第60 d, SO2处理组果实蔗糖、葡萄糖和果糖含量较对照组有不同程度的提高。SO2通过上调细胞壁转化酶(Cell wall invertase, CWINV)、中性/碱性转化酶(Neutral/Alkaline invertase, CIN)、蔗糖合成酶(Sucrose synthase, SuSy)和蔗糖磷酸合成酶1(Sucrose phosphate synthase1,SPS1)基因的表达水平来维持葡萄果实的高糖含量。调控己糖激酶2(Hexokinase2,HxK2)、果糖-二磷酸羧醛酶(Fructose-bisphosphate Aldolase, FBA)、甘油醛-3-磷酸脱氢酶(Glyceraldehyde-3-phosphate dehydrogenase, GAPDH)和磷酸甘油酸激酶(Phosphoglycerate kinase, PGK)基因的表达水平,SO2激活葡萄果实采后贮藏过程中的光合活性。【结论】SO2可以调控相关基因的表达水平以调节木纳格葡萄采后贮藏中的糖代谢相关途径。【Objective】To study the effect of sulfur dioxide(SO 2)on sugar content and sugar metabolism pathway in Munage grape fruit.【Methods】The changes of sucrose,glucose and fructose contents in Munage grapes during storage were determined by high performance liquid chromatography(HPLC)and simultaneously,the expression differences of 20 genes related to glucose metabolism were evaluated.【Results】SO 2 treatment significantly increased the glucose content from 0 to 10 days of storage.On the 60th day of storage,the contents of sucrose,glucose and fructose in the SO 2-treated grape were increased to varying degrees compared with that of the control group.SO 2 up-regulated cell wall invertase(CWINV),neutral/alkaline invertase(CIN),sucrose synthase(SuSy)and sucrose phosphate synthase1(Sucrose phosphate synthase1,SPS1)gene expression level to maintaine high sugar content in grape during storage.Moreover,SO 2 activated the photosynthetic activity of grape by inducing the expression of hexokinase 2(HxK2),fructose-bisphosphate Aldolase(FBA),glyceraldehyde-3-phosphate dehydrogenase(GAPDH)and phosphoglycerate kinase(PGK).【Conclusion】SO 2 can regulate the expression levels of related genes to modulate the glucose metabolism-related pathways in the postharvest storage of Munage grapes.
