摘  要:目的 探究超声微泡(UM)介导miR-187-3p对乳腺癌细胞增殖、迁移和侵袭的影响及可能的作用机制。方法qRT-PCR检测不同乳腺细胞中miR-187-3p表达水平。双荧光素酶报告基因实验验证miR-187-3p与S100钙结合蛋白A4(S100A4)的靶向关系。使用miR-187-3p mimic、miR-NC或UM介导的miR-187-3p mimic、miR-NC处理MDA-MB-231细胞,并过表达S100A4进行回复实验,采用qRT-PCR、Westernblot检测细胞中miR-187-3p、S100A4mRNA和蛋白表达水平,CCK-8、EdU染色检测细胞增殖能力,划痕愈合实验、Transwell实验检测细胞迁移、侵袭能力。结果 miR-187-3p在人乳腺癌细胞中低表达,其中MDA-MB-231细胞中miR-187-3p表达水平变化最为明显。过表达miR-187-3p可抑制MDA-MB-231细胞增殖、迁移和侵袭;UM介导的miR-187-3p进一步增强了miR-187-3p对MDA-MB-231细胞增殖、迁移和侵袭的抑制作用;经验证,MDA-MB-231细胞中miR-187-3p与S100A4存在靶向调控关系,且过表达S100A4可部分减弱UM介导的miR-187-3p对MDA-MB-231细胞增殖、迁移和侵袭的抑制作用。结论 UM介导的miR-187-3p可能通过负靶向调节S100A4抑制乳腺癌细胞增殖、迁移和侵袭。Objective To explore the influence of ultrasound microbubble(UM)-mediated miR-187-3p on the proliferation,migration and invasion of breast cancer cells and its possible mechanism.Methods The expression level of miR-187-3p in different breast cells was detected by qRT-PCR.The targeting relationship between miR-187-3p and S100 calcium binding protein A4(S100A4)was verified by dual-luciferase reporter gene assay.MDA-MB-231 cells were treated with miR-187-3p mimic,miR-NC or UM-mediated miR-187-3p mimic,miR-NC,and S100A4 was overexpressed for recovery experiments.The mRNA and protein expression levels of miR-187-3p and S100A4 in cells were detected by qRT-PCR or Western blot.Cell proliferation ability was detected by CCK-8 and EdU staining.Cell migration or invasion ability was detected by scratch healing assay and Transwell assay.Results The expression of miR-187-3p was more lower in human breast cancer cells,and the expression level of miR-187-3p in MDA-MB-231 cells changed most obviously.Overexpression of miR-187-3p could inhibit the proliferation,migration and invasion of MDA-MB-231 cells.UM-mediated miR-187-3p further enhanced the inhibitory effects of miR-187-3p on the proliferation,migration and invasion of MDA-MB-231 cells;it had been verified that there was a targeted regulatory relationship between miR-187-3p and S100A4 in MDA-MB-231 cells,and overexpression of S100A4 could partially attenuate the inhibitory effects of UM-mediated miR-187-3p on the proliferation,migration and invasion of MDA-MB-231 cells.Conclusion UM-mediated miR-187-3p may inhibit the proliferation,migration and invasion of breast cancer cells by negatively targeting S100A4.
