摘  要:目的通过对不明原因复发性流产和正常早期妊娠进行外周血WNT4基因突变位点筛查,从而研究WNT4基因(人外周血中Wnt4基因)突变是否与复发性流产有关。方法对100例不明原因复发性流产和100例正常早期妊娠进行外周血WNT4基因突变位点筛查,查找WNT4基因编码序列是否存在基因突变,从而研究WNT4基因突变是否与复发性流产有关。结果观察组及对照组研究对象WNT4基因的第一、三、四、五外显子PCR产物测序结果与正常序列相比较未发现明显突变。观察组2例患者存在已知的WNT4基因第二外显子SNP(单核苷酸多态性rs16826648),观察组基因型频率与对照组差异无统计学意义(P>0.05)。结论WNT4基因突变与复发性流产可能无明显相关性。Objective To screen for peripheral blood WNT4 gene mutation sites in unexplained recurrent miscarriage and normal early pregnancy,in order to investigate whether mutations in the WNT4 gene(WNT4 gene in human peripheral blood)are associated with recurrent miscarriage.Methods A total of 100 cases of unexplained recurrent miscarriage and 100 cases of normal early pregnancy were screened for mutations in the WNT4 gene in peripheral blood to determine whether there were genetic mutations in the coding sequence of the WNT4 gene,and to investigate whether the WNT4 gene mutation is related to recurrent miscarriage.Results The sequencing results of the first,third,fourth and fifth exon PCR products of WNT4 gene in the observation group and the control group were not significantly mutated compared with the normal sequence.Two patients in the observation group had known SNPs in the second exon of the WNT4 gene(single nucleotide polymorphism rs16826648),and there was no significant statistical difference in genotype frequency between the observation group and the control group(P>0.05).Conclusion There may be no significant correlation between WNT4 gene mutations and recurrent miscarriage.
